HPTLC INSTRUMENTATION OVERVIEW
ABSTRACT
HPTLC is one type of planar chromatography.
Now a day HPTLC is indispensable of analysis in the natural products. It has been preferred over TLC and HPLC.
Because HPTLC is independent of sample application, chromatogram development, detection, etc., it is not only
instrumental TLC but entire concept that include widely standardize methodology based on validated method.
This type of analysis is controlled by Win CATS and Vision CATS software’s. The present report is helpful to carrying
out analysis of phytochemicals (analytes) present in the herbal drugs or natural products.
In this project work, we report a review on HPTLC of various analyses so that the students get compilation of the
analysis of phytochemicals reported in the reputed literature. The literature suggests that some of the phytochemicals can
be analyzed under UV light of short (254 nm) and long (366 nm) wavelengths for e.g., ursolic acid, lupeol, withferin A,
andrographolide, oleinolic acid, eugenol, ferolic acid, B sitosterol etc.
The literature also include the applications of HPTLC and its methodology. It include a brief introduction of
chromatograms. The tabulated data on the profiles of the phytochemicals analysis include acuracy, precision, sensitivity
(LOQ, LOD), linearity range, linear regression equation, Rf value, robustness. It also reports use of different spraying
reagents used in HPTLC analyses.
INTRODUCTION
Planar Chromatography as opposed to column chromatography (e.g. GC, HPLC) utilizes a flat
(planar) stationary phase for separation.
HPTLC is an entire concept that includes a widely standardized methodology based on scientific
facts as well as the use of validated methods for qualitative and quantitative analysis.
The most advanced form of instrumental TLC is commonly called high performance thin- layer
chromatography (HPTLC), but the term does not simply imply instrumental TLC on special high
performance layers.
The possibility of visual evaluation ofseparated samples on the plate is one of the most valuable
aspects of TLC.
PRINCIPLE
The HPTLC works on the same principles as TLC such as the principle of separation is
adsorption.
The mobile phase or solvent flows through the capillary action.
The analytes move according to their affinities towards the stationary phase (adsorbent).
The higher affinity component travels slower towards the stationary phase.
HPTLC METHODOLOGY
Difference between TLC and HPTLC
