Copper sulphate method also
known as specific gravity method
is a method of analysis of Hb
based on specific gravity where it
is assumed that donor have the
normal protein levels.
Interpretation
In this method a drop of blood is
allow fall on copper sulphate
solution of specific gravity 1.053
from a hight of 1cm. Specific
gravity of 1.053 is equal to the
12.5 gram of Hb. Now if the drop
of blood is float on the surface of
copper sulphate solution, then it
means the Hb level is low
whereas if the drop of blood get
sink on bottom of the solution,
then it's simply means that the
hb is high.

Copper Sulphate method.
• This method is based on the estimation of specific
gravity of blood, assuming that the donor has
normal protein levels.
• It is a qualitative analysis.
• It is completely manual method which involves
preparation of stock solution first and then we have
to visually analyse the dispersion.
• For interpretating the result, we must wait for
around 15 seconds.
• This test can give false negative if it is not performed
carefully common reason for false negative are
formation of air bubble , dropping of blood from
inadequate hight.
• It also may lead to the false high value of hb as
because sometime copper also react with the other
proteins other than hb cause dispersion of blood .
• Reagent solution should be change after 25 test as it
effect the specific gravity of the solution.
