PENICILLIN PRODUCTION

BY
SOLID STATE FERMENTATION
Prepare spore inoculum, Penicillium chrysogenum strain in PDA medium flask,
incubated at 27°C, for one week,
suspend the spores in sterile water, inoculum size 0.5 × 106 /ml

Prepare raw material, Sugarcane bagasse free of sugars from sugar factory,
pass through 30-50 mesh size,
moisten to 50%, sterilize in autoclave

To the raw material, add media components (2×) and spores, adjust pH.
Corn Streep liquor 30 g/L, Lactose 30 g/L, Glucose 5 g/L, Calcium carbonate 3 g/L,
Lard oil 1.87 g/L, Phenyl-acetamide 0.05 g/L, Silver nitrate 3 g/L, Zinc sulphate 0.044 g/L,
Magnesium sulphate 0.25 g/L, pH 5.5

Dry the fermented material at 60°C for 48 hrs.

Mix 3 mL Ethyl acetate

Centrifuge 10 min, 5000 rpm
