Test to distinguish monosaccharide / disaccharide.
Principle : oxidation of reducing sugar by copper ion to form a carboxylic acid and copper oxide, results in formation of red Coloured precipitate. Procedure : 1 ml of sample + 2 ml of barfoed’s solution , boil in water bath for 1 min , red color/precipitate is formed. Monosaccharide: result in 1-5 minutes Disaccharide : result in 7-9 minutes. Also to check the presence of aldehyde /ketone. Reagent : alkaline ammonical silver nitrate Procedure : take two dry test tubes and add 1 ml of sample in one test tube and 1 ml of distilled water in another as blank + 2ml of tollen’s reagent to both test tubes and keep both the test tubes in water bath for 1 min Observe the silver mirror on sides of the test tubes indicates the presence of aldehyde.
