Blood collection and

technique
Blood collection = is the process of taking blood sample from patients or subject.
This process involved two method
Capillary blood collection
Venous blood collection
• Capillary blood collection/puncture = is frequently use when small
quantities of blood is required or needed, for example HB,PCV, GLCOUSE
TEST, WBC/RBC, OR BLOOD SMEAR Preparation.
• Site to be puncture
• Adult = the ring finger or middle finger
• Children = the ear lobe
• Infant = big toe or kneel
• Apparatus
• Cotton wool
• 70% alcohol
• Lancet
Techniques
• Rub site vigorously with cotton moistened with alcohol.
• Allow the skin to dry, make a puncture 2-3 mm deep with the lancet.
• Stop the blood flow by applying slight pressure with cotton wool at the
site.
• Advantage of capillary blood collection/puncture .
• It is obtained with ease.
• It is pure no anti-coagulant is added.
• Disadvantages of capillary blood collection /puncture.
• Only small amount of blood can be obtained.
• Platelet count can not be performed.
• VENOUS BLOOD COLLECTION /PUNCTURE= Venous blood is the
withdrawing of blood sample from the vein, and is used for most test that
require larger quantities of blood, plasma, or serum.
• SITE TO BE PUNCTURE = the three main veins of the fore arm, ante-cubital
fossa of the arm
• Apparatus
• Syringe and needle
• Tourniquet
• Cotton wool
• 70% alcohol sample bottle (EDTA)
PROCEDURE
• Assemble the syringe/needle.
• Allow the subject to sit comfortable.
• Prepare the arm by swabbing with cotton moistened with 70% alcohol.
• Apply a tourniquet 6-7m above the elbow.
• Select a vein.
• Penetrate the skin first and then the vein, tourniquet should be released
the moment blood start entering the syringe.
• Apply a ball of cotton wool to the puncture site and gently withdraw the
needle.
• Cover the needle with it’s cap remove it from the nozzle of the syringe
then empty the blood into a sample bottle.
• Re-inspect the venipuncture site .
Advantage of venous blood
collection/puncture
• It provide sufficient amount of blood
• May be frozen for future use
• it has plasma and serum

• DISADVANTAGE OF VENOUS BLOOD COLLECTION/PUNCTURE

• It is technically difficult in children
• It can lead to hematoma
Assignment
• 1) what is the difference between capillary and venous blood
• 2)state the meaning of the following and their function
• EDTA
• TRI-SODUM
• HEPARIN
• DOUBLE OXALATE
• State the different between plasma and serum
