    BCA Protein Assay

    Procedure 1 (standard assay ) – test-tube ( Sample : WR = 1 :20 )

    • Reagent A: → 1g BCA + 2g Na-carbonate + 0,4g NaOH + 0,95g Na-bicarbonate + Na-tartrate


    → fill up to 100 ml with dH2O
    → echilibrate pH = 11,25 ( 10M NaOH )
     Reagent
    • Reagent B: 0,4g cupric sulfate in 10ml dH2O

    • Standard working sol. (SWR): 100x ReagentA + 2x ReagentB ( 50:1 )

     Assay

    2,0ml WR

    Incubate cool
    Read ( 562nm )
    30 min, 37C 10 min, RT
