SOP for assessing efficiency of a chromatographic peak with TotalChrom software.
1. First, you must of course perform a chromatographic run in TotalChrom.
2. In TotalChrom Navigator (the main window), Click on the Graphic Edit button. 3. The window Graphic Method Editor appears. 4. Open your run in this window by: 5. File 6. New Data File... 7. Click on the run of interest in the TotalChrom File-Open window. Click Open. 8. Now your chromatogram of interest should have appeared in the window. 9. Ensure that the baseline is drawn correctly under the peaks of interest. If they are not, click on the Manual Integration button (fifth icon from the right). 10. Draw the baseline correctly with the mouse (left-clicking during drawing). 11. If not drawn correctly, delete the drawn baseline by right-clicking on the baseline and choosing Delete Baseline... 12. Repeat until satisfied. 13. Click the Edit components icon (fourth from the right) 14. Click on your peak of interest, and name it in the right side of the window. 15. Choose Next. 16. Click on the Return button (top left of screen). 17. Choose File 18. Save Result File As... 19. OK
20. Save file in an appropriate site in the TotalChrom File-Save-As window.
21. Close the Graphic Method Editor (dont save Modified Method) 22. In the TotalChrom Navigator window, click on Apps in the top of the window. 23. Choose Suitability. 24. Choose File 25. Data 26. Add 27. (add your file) 28. OK 29. Choose File 30. Print Preview... 31. In Print Options, choose Plot and report 32. Click OK. 33. Check that your peak of interest has been treated. 34. Print pages. 35. Take a break after this traumatic computer task.
