o TIR1-axr
 SCF protein binds AUX proteins

 If you add auxin, ARF becomes free


 If have SCF mutant… ARF will NOT be released, KEY POINT
 Mutant situation, aux never binds to SCF-Tir1
 Wildtype: binds in presence of auxin
 TIR1 refers to a wildtype gene that acts as a SCF complex
component that normally degrades AUX proteins in presence of
auxin (wildtype TIR1 protein)
 Mutant = tir1-, the SCFTIR1 is inactive and can no longer bind to
the aux protein
• If you can’t bind aux, you can’t respond to auxin
 Left side, wildtype, right side mutant
• Mutant is auxin RESISTANT
