(1). Bioprospecting (MBTD/PATH/01)
Two compounds, i) steroidal and ii) substituted unsaturated fatty acid types have been obtained from brown
seaweed Sargassum longifolium after repeated chromatographic purifications
The preliminary nature of compounds has been revealed from their spectral data
The compounds showed broad spectrum antibacterial activity against pathogens of aquaculture importance
namely, Vibrio parahaemolyticus, V. vulnificus, V. harveyii, Pseudominas fluorescens and Aeromonas
hydrophilla when tested with their DMSO solutions
Seeds (140g) have been obtained from dried plants (1250g) to find its further use
Alcoholic extract (18g) of young plants (500g) reported to have antioxidant and hypocholesterolemic
properties was obtained
Nutrient Profiling of fish and fish feeds: (April 2009 – March 2010)
HPLC instrument was upgraded with accessories of column oven and fluorescence detector
Calibration of HPLC instrument with vitamin standards A, E, D2, D3 and essential fatty acid standards, DHA,
DPA, PUFA and FAME (Sigma)was done with their respective RTs
Required fish samples were procured for the analysis
Fish specimens of sardines, ribbon fish, grouper and silverbellies were procured for analysis.

(v). Administration/Management/Coordination
Apart from research project work, I was also given various administrative works, viz. In-Charge of the Centre during
the absence of regular Scientist-in-Charge on 25/5/2009, 30/5/2009, 26/6/2009, 16/7/2009, 4/8/2009, 23/3/2009;
DDO on 26/5/2009; Co-signatory for cheques on 10/7/2009.
I was also in-charge of the central block lab and occasional Library-in-Charge during the concerned official’s
absence.
I had been given the In-Charge of Guest House to ensure its smooth functioning during the visits and the stay of the
ICAR dignitaries and other officials visiting on official tours. Daily transactions with respect to purchases for the guest
house (grocery and other consumables), Daily accounting of sales and proceeds, Guest house room rent collection
and its remittance to ICAR A/c, labour management for guest house were routinely done during 2009-1010.
Food and refreshment work was managed i). For the participants for 10 days during “Marine Ornamental Fish Culture
Training” conducted during 12 t0 21 October 2009;
ii). For the participants during Marine Ornamental Fish Culture Training sponsored by GOMBRT from 8/2/2010 to
18/2/2010

Detailed Report
Consequent on my transfer from Headquarters, Cochin to Mandapam, I joined Mandapam Regional Centre in April
2009. The lab with minimum available facility was set up using the indented items of lab equipments, chemicals and
glassware and the lab equipments transported from Head quarters, Cochin.
1). In-house project: i). MBTD/PATH/01- Pathogen profiling, diagnostics and health management
Inmaricultured finfish and shellfish
ii) PNP/NUT/01- Formulation and evaluation of larval and growout feed for marine
crabs, lobsters, ornamentals and cage farmed fin fish
2). Outreach project: Nutritional profiling and evaluation of fish as dietary component
MBTD/PATH/01: Under this project the following works were carried out.
Two pure compounds obtained from brown seaweed, Sargassum longifolium on preliminary spectral data revealed
the steroidal and substituted unsaturated fatty acid nature. The steroidal compound on repeated NMR analysis
showed the basic unit of fucosteryl moiety with observed M+1 data of mass spectrum. The fatty acid compound is
expected to have a cyclopropyl substituted side chanin moiety as observed in its mass spectra.
Bio assay of these two compounds showed antibacterial activity against aquaculture pathogens of Vibrio
parahaemolyticus, V. vulnificus, V. harveyii, Pseudomonas fluorescens, Aeromonas hydrophilla with inhibition range
from 6.3 to 8.4 mm when their DMSO solutions were screened.
Bioprospecting:
The halophyte, Salicornia brachiata available in Mandapam coasts as a promising alternative livelihood source of the
coastal communities was explored. Young plants were collected during May-June 2009 from Thondi coast. Matured
and dried plants were collected from Mandapam coast during August 2009. Ethanol extract (18g) of the shade dried
and powdered young plants (500g), was obtained as a dark green residue. This is reported to have medicinal
properties like antioxidants, hypocholesterolemic, etc. Matured plants (1.25kg) were sundried and processed to get
140g of seed (11% ) as reported in the literature. Oil extraction and its analysi would reveal the nutritional value with
reference to essential fatty acids.
The semi preparatory grade HPLC (Shimatzu) of the lab was upgraded with additional essential accessories of
i). Column oven and ii) Fluorescence detector in June 2009. The overall functioning of the instrument was
streamlined. Demonstration with caffeine standard was done in August 2009. Fresh standards (Sigma) of vitamins A,

E, D2, D3 and essential fatty acids of DHA, DPA, PUFA and FAME were procured in November and standardized
with their respective RTs in December 2009. Necessary fish specimens of sardines, ribbon fish, grouper and
silverbellies were procured for the analysis of their vitamin content.
Outreach project:
HPLC instrument has been upgraded and put to normal working conditions for the analysis of dietary fishes selected
from sardines, ribbon fishes, grouper and silverbellies. Rectification of the UPS of the instrument is awaited for the
periodical analysis of the specimens.
Proposal was made for centre’s mariculture work on cobia during the project review meeting on 6th January 2010
2. Workshops attended
i) Hindi work shop was attended on 22/3/2010 conducted by Asst. Director (OL)
ii) Work shop on Breeding and Seed Production of Marine Fin fishes with special reference to Cobia was attended on
30/3/2010 organized by NAIP at the Centre
iii) Marine Fisheries Census work shop was attended on 17 th & 18th Marth 2010 for the census to be conducted
during April- May 2010.
A paper entitled “Antibacterial and antifungal principles from Southeast Indian Coast sponges” has been submitted
for consideration for publication to JMBAI on 18/12/2009.

Report on surprise inspection of cash for the quarter April to June, 2010
The surprise inspection was done at 12.30 hrs on 03/07/2010 to verify the cash balance and the
transactions under various heads.
1. The sum total of the cash in hand as per the cash book was found to be Rs.2,41,737/-
(Rupees two lac fortyone thousand seven hundred thirty seven only) tallying with the
figure of the cash book.
2. The cash on hold with respect to thrift society of Kochi & Chennai, CTD, senior club
was found to be Rs.22,037/- (Rupees twenty two thousand thirty seven only)
3. Certification was made after verification of the transactions in the cash book.
