Kidney Tumor Necrosis Factor Alpha (TNF-) in Type-2 Diabetes Mellitus Rat Model

ABSTRACT

Background: Nephropathy diabetic was type-2 diabetes mellitus complication that were mediated by
inflammation state. This condition would activated mesangial cell to secreted kidney TNF-. Nipah
leaves midrib (Nypa fruticans Wurmb.) contains flavonoid, tanin, polyphenol and alkaloid that act as
antidiabetic, antioxidant and anti-inflammation. This study aim to know the effect of nipah leaves
midrib (Nypa fruticans Wurmb.) on the inhibition nephropathy diabetic complication by observing
kidney TNF- level in type-2 diabetes mellitus rat model.
Methods: This in vivo study used control group post test only design with male Rattus novergicus.
Diabetic rats were induced by High Fructose Diet (HFD) and single dose Streptozotocin (STZ) 35
mg/kgbw via intraperitoneal. The administration of nipah leaves midrib decocta with the dose of 200,
400 and 800 mg/kgbw were given each orally for 30 days. Kidney TNF- were measured in ELISA.
The data was analyzed using one way ANOVA continued by LSD test (p<0.05).
Results: TNF- level in negative control group and positive control group 659,4 18,5a pg/mL and
767,4 30,2b pg/mL respectively while in type-2 diabetes mellitus group that was administrated by
nipah with the dose of 200, 400 and 800 mg/kgbw 731,4 15,5c pg/mL, 685,4 9,9a pg/mL and 579,9
16,3d pg/mL respectively. In all groups that was administrated by nipah leaves midrib decreased
TNF- level were decreased compared to negative control and positive control.
Conclusion: The administration of nipah leaves midrib decocta with the dose of 200, 400 and 800
mg/kgbw were able to decrease kidney TNF- level approximately 5%,11% and 24% respectively
compared to positive control group (p<0.05).
