Preparation of Quinine sulphate stock solution 1000ppm
Weigh accurately 0.12707g of Quinine sulphate dihydrate and transfer it into 100ml of volumetric flask. Pipette 5ml of 0.1M sulphuric acid into flask. Dissolve quinine sulphate completely before diluting to volume. Preparation of intermediate Quinine stock solution 10.0ppm Pipette out 5ml of 100ppm solution in 500ml of volumetric flask add 250ml of 1M sulphuric acid dilute to volume with distilled water. Preparation of Quinine standard solution: Using graduate pipette cold 1.0, 3.0, 5.0, 7.5 and the 10ppm intermediate stock solution into properly labelled and as 100ml volumetric flask. The result in standard solution of 0.1, 0.3, 0.5, 0.75 and 1.0ppm. 0.05M Sulphuric acid used as blank. Preparation of Quinine unknown: Unknown solution is also prepared in same manner as that of standard Quinine solution and fluorescence intensity was observed.
