Aim: To observe and draw……………………………

Apparatus and Materials: 1) Microscope

2) name of slide
Method:
1. The microscope was switched on and the platform lowered by moving the rough
focus knob.
2. The lens was set to the lowest magnification (x4) by moving the nosepiece.
3. The slide was placed on the platform and secured with the clip.
4. The platform was slowly raised until the specimen comes in focus.
5. The fine focus knob was used to bring the specimen into sharp view.
6. The platform knob was used to find items on slide.
7. Steps 2-6 were repeated with magnification x10 and x40.

Results: table and drawings

PRECAUTIONS

1. The microscope was held at the neck and the base while transporting to avoid damage
2. The slides were held at the edges to avoid getting fingerprints on the glass.
3. A break was taken from looking at the microscope to avoid eye fatigue.
4. The stage was lowered before changing the lens to avoid hitting and damaging the slide.
5. Two hands were used when using the coarse and fine focus to avoid damaging it.

SOE

1. Parallax error when reading eyepiece graticule to get measurement of cell, because
eyepiece graticule was not read at eye level.

LIMITATIONS

 The wavelength of light which illuminates the specimen limit the resolution as the
wavelength of light ranges from 400nm to 700nm, the microscope cannot resolve parts of
the specimen that are closer than 200nm, so it was difficult to differentiate between
structures.
 Light has a relatively long wavelength, and this limits the microscope to magnify by a large
amount without distorting the image making it difficult to see small structures.
