Preparation of Benzoyl Glycine (Synthesis of an Amide)

Materials Required

Procedure
1. In a conical flask, 8 ml of 10% of NaOH solution.

2. Dissolve 1.5 g of glycine in the flask.

3. Add 1.5 ml of Benzoyl Chloride.

4. Shake the flask well for 5 minutes.

5. Add sufficient crushed ice, followed by concentrated HCl which is added in excess,
dropwise
6. Precipitation will occur.

7. Filter under suction.

8. Dry and weigh

Calculation

Here limiting reagent is glycine; hence yield should be calculated from its amount.

Molecular formula of glycine = C2H5NO2

Molecular formula of benzoyl glycine = C9H9NO3

Molecular weight of glycine = 75 g / mole

Molecular weight of benzoyl glycine = 179 g / mole

Theoretical Yeild

75 g of glycine forms 179 g of benzoyl glycine

Therefore, 1.5 g of glycine will form x grams of benzoyl glycine

X= ( 179 x 1.5 ) / 75 = 3.58 g

Practical Yeild = 2.43 g

% Yeild = (Practical Yeild) / (Theoretical Yeild) x 100

= (2.43 / 3.58) x 100

= 67.87 %

Application

Hippuric Acid is an acyl glycine produced by the conjugation of benzoic


acid and glycine, found as a normal component in urine as a metabolite of
aromatic compounds from food. Increased urine hippuric acid content may
have antibacterial effects.

Hippuric acid can be used to study cell biology, chemical biology, bioactive
small molecules, amino acid derivatives, peptide synthesis, chemical
synthesis and nutrition. Hippuric acid has been used to inform the
metabolism and urinary excretion of procyanidins.
