From the graph peak height vs concentration, using the equation Y=mx+c to determine the
original concentration.

y=1102.2 x +28.518
To find the original concentration, the value of peak height at 0 concentration of standard
caffeine was used in this calculation which is 24.79.

24.79=1102.2 x +28.518
x=−0.00338 mg/mL
The value -0.00338 is for after 50 mL dilution concentration. So, the original concentration is

original concentration=−0.00338× 10=−0.0338 mg/ mL


Therefore, to calculate original concentration for peak area will use the same calculation.

y=16742 x+ 503.3
408.3=16742 x +503.3
x=−0.00567 mg/mL
original concentration=−0.00567× 10=−0.0567 mg/mL
