26. Study the effects of temperature on the activity of yeast.

OBJECTIVE: To investigate the effect of temperature on the rate of anaerobic respiration in


yeast.
PROBLEM STATEMENT: How does the temperature affect the rate of anaerobic respiration
in yeast?
HYPOTHESIS: The higher the temperature, the faster the rate of anaerobic respiration in yeast.
MANIPULATED VARIABLE: The temperature
RESPONDING VARIABLE : Rate of anaerobic respiration
CONTROL VARIABLE : Volume of yeast suspension

APPARATUS AND MATERIAL :Boiling tube,manometer,stopwatch,marker,ruler,water


bath,rubber stopper,rubber tubing,retort stand,measuring
cylinder,yeast suspension,glucose solution,paraffin oil.
PROCEDURE:
1. Set up the apparatus.
2. Place the boiling tube with 15cm³ of glucose solution in a beaker of ice for 5 minutes.
3. Add 15cm³ of yeast suspension into the boiling tube.
4. Mark the initial height of the coloured liquid in the manometer.
5. Start the stopwatch and measure and record the level of the coloured liquid in the
manometer after 10 minutes.
6. Record all the data in a table.
7. Repeat steps 1 to 5 by placing the boiling tube in water baths at temperature of 20°C
40°C and 60°c.
8. Make sure all the joints of the apparatus are airtight and repeat the experiment twice at
each temperature to get an average reading.
9. Calculate the rate of anaerobic respiration by using the formula:

CONCLUSION: The higher the temperature,the higher the rate of anaerobic respiration in
yeast.
