When you want to agree with an affirmitvie remark, use: yes + pronoun + the auxiliary
John: It’s hot today. Rachel: Yes, it is.

When you would like to agree with a negative ramark, use: no + pronoun + the negative auxiliary
John: It isn’t cold today. Rachel: No, it isn’t.

To disagree with affirmative or negative remarks, use: (oh) no + pronoun + the auxiliary
(the affirmative auxiliary if the first verb is negative, the negative auxiliary if the first verb is affirmative)
John: It’s hot today. Rachel: (Oh) no, it isn’t.
John: It isn’t cold today. Rachel: (Oh) yes, it is.
