B. What is the correct position of plated medium during incubation of culture?

During storage? Explain your answer

The plated medium or the Petri dishes with culture medium must be stored and

incubated INVERTEDLY, Because of the following reasons:

1. When the plates are incubated in the normal position, water evaporation from

media occurs, these vapors condense on the lid of the Petri dish thus will drop

fall on the colonies grow on the media surface. This will cause a mix-up colony

and spread throughout the plate surface.


2. The water evaporation from media can cause dryness that can affect the

Microbial growth. Therefore, petri dish with culture media can also be stored for a

longer period in an inverted position

3. The lid of the Petri dishes may contain any contamination that spreads on the

media and grows together with the sample organism,

4. Accidentally opening of the petri dish may be avoided if the petri dish is

incubated and stored inverted.
