PRACTICAL ASSIGNMENT 2: BLOOD GLUCOSE TEST

201705714
1. What are the principles of blood glucose test (6)

Glucose oxidase catalyse the oxidation of Beta D-glucose present in the plasma to D-
glucono-1,5-lactone with the formation of hydrogen peroxide; the lactone is then slowly
hydrolysed to D-gluconic acid. The hydrogen peroxide produced is then broken down to
oxygen and water by a peroxidase enzyme. Oxygen then react with an oxygen acceptor such
as ortho toluidine which itself converted to a coloured compound, the amount of which can
be measured colorimetrically.

2. Define glucose oxidase (2)

an antibacterial flavoprotein enzyme, obtained from Penicillum notatum and other fungi,
which is antibacterial only in the presence of glucose and oxygen, its effect being due to the
oxidation of d-glucose to d-glucono-δ-lactone, with the coconversion of O2 to H2O2; used in
the preservation of food and in assays for glucose levels.

3. What role does glucose oxidase have in blood glucose test? (2)

Glucose oxidase (GOD) catalyzes the oxidation of glucose to gluconicacid. The formed
hydrogen peroxide (H2O2), is detected by a chromogenic oxygen acceptor, phenol-
aminophenazonein the presence of peroxidase (POD).
