Factors that affect Enzyme Activity

Introduction

Enzymes are proteins that catalyze various biochemical reactions. Their activity is affected by various
factors such change in enzyme concentration, change of substrate, temperature, and pH.

In this activity, you will investigate the effect of change in pH and temperature on the enzyme itself.

Methodology

Select any of the following, depending of availability, lettuce, apple, potato, or eggplant.

For the Change in temperature:

1. Cut the vegetable of your choice into three.


2. Put one portion at room temperature. Soak the other portion in boiling water for 2 minutes.
After 2 minutes, remove from boiling water and leave at temperature.
3. Put the last portion inside the fridge. This option is for those who went home and who have
fridge.
4. Record the change in color for every 2 hours for 10 hours.

Change in pH

1. Again, cut small piece of vegetable of choice into two.


2. On to one portion, apply generous amount of lemon juice or calamansi juice or vinegar on its
surface.
3. Apply only water on the other half.
4. Record the change in color for every 2 hours for 10 hours.

-Make a report on what you have observed between the two experiments that you performed. You
may include the pictures.

- What is the enzyme that is responsible for discoloration of the plant stuff?

-Discuss the effects of the change in temperature and pH on the enzyme

- Short reflection.
