ABSTRACT
Oral hygiene that is not properly maintained will cause diseases in the oral cavity. The common periodontal diseases are
including gingivitis and periodontitis. Streptococcus is a heterogeneous, gram-positive and round-shaped bacterium that
typically forms pairs or chains during its growth period. Among Streptococcus bacteria, Streptococcus pyogenes is
known as pathogenic microorganisms that are associated with various oral cavity diseases. Various traditional
medicines have been used by the community to treat various infectious diseases caused by bacteria and fungi, one of
which is the brotowali stem (Tinospora crispa L Miers). Since brotowali has active tannins, flavonoids, saponins,
alkaloids that are scientifically proven to have the ability to be antimicrobial, antinociceptive, anti-inflammatory,
antiproliferative and antioxidant. The purpose of this study was to determine whether the antibacterial effects of
brotowali stem extract (Tinospora crispa L. Miers) on the growth of Streptococcus pyogenes. This study is a laboratory
experimental with post-test only control group design, using Mueller Hinton Agar (MHA) media testing. Tests were
carried out using the Kirby-Bauer method at concentrations of 25%, 50%, 75%, 100% and negative control of 96%
ethanol and positive control of vancomycin. Observations were made after 24 hours of incubation period. The results of
the analysis in this study note that the optimal concentration of the treatment given to this bacterium is the brotowali
extract at 50% concentration. Brotowali stem extract (Tinospora crispa L. Miers) has the activity of inhibiting the
growth of Streptococcus pyogenes bacteria, where there are significant differences between the control group and the
treatment group. This shows that the antibacterial activity of brotowali (Tinospora crispa L. Miers) stem against the
growth of Streptococcus pyogenes.
