Hints for Hill Reaction Lab

Title- The Hill reaction in isolated chloroplasts


Aim: List about 3 main activities that were done.
Introduction:
● General description of photosynthesis:
Show chemical equation summarising process, location and maybe a
drawing of the chloroplast
● Focus on hill rxn: Light/Dark rxn.
● Description/Principle behind method
● Z-scheme
Method: Same point as previously stated in other labs.
Results:
● Part A. Determination of chlorophyll content.
Calculation of total yield - Using A = Ecl
1 ml of diluted chloroplasts contains 0.0025 mg chlorophyll
10 ml of diluted chloroplasts contains 0.025 mg chlorophyll
0.1 ml stock contains 0.025 mg chlorophyll
1 ml stock contains 0.025/0.1x 1 = 0.25mg/ml (concentration)(1 mk)
NB: vol. (mLs) is variable cause of resuspended pellet + vol pellet was
resuspended in, taken from manual.
Yield
1 ml stock contains 0.025mg
(Vol. total stock you measured Y) contains:-
0.025 x Y = Z mg chloroplast/chlorophyll
Assuming wet weight of plant tissue was 20 g,
Total yield = Z/20 = W mg/g of plant leaves
