MATERIALS AND METHODS

Preparation of Standards
In this experiment, 2.5 mL of 1000-ppm standard of F- was diluted to 250 mL in a
volumetric flask to obtain a 10-ppm solution. From the 10-ppm solution, 5, 10, 25,
50, and 75-mL aliquots were transferred to 100 mL volumetric flask to obtain 0.5,
1.0, 2.5, 5.0, 7.5 ppm standard, respectively. Then, 1 mL of TISAB (Total Ionic
Strength Adjustment Buffer) solution was added, and solution was diluted to mark
with distilled water.
Preparation of Sample for Fluoride Analysis
*Mouthwash
Briefly, 1 mL of mouthwash (insert brands) was added on each of the 100-mL
volumetric flask and added with 1 mL of TISAB solution, then diluted to mark
using distilled water.
*Toothpaste
About 2.0 g of sample toothpaste (brand) was weighed and transferred to 100 mL
beaker, added with 1 mL of TISAB solution and 50 mL distilled. Then, the mixture
was boiled until the toothpaste was completely dissolved in the solution. The
solution is then cooled and transferred into a 100 mL volumetric flask and diluted
to mark using distilled water.
Instrument Set-up and Operation
Analysis of Fluoride
After setting-up the _____, the electrode was immersed to the 0.5-ppm standard
which was transferred to a 100-mL beaker. It is then stirred manually until a
constant value for the potential (mV) was obtained. The process was repeated for
all concentration of the standards. Then, the measured potential (mV) is plotted
against the log of concentration (ppm) of the standards to obtain a slope which
should have a value of -59.4± mV indicating a normal operation.
For the analysis of fluoride in mouthwash, 30 mL of the unknown solution was
transferred to a 100-mL beaker and the electrode was immersed and the
potential was recorded. The concentration of fluoride in mouthwash was then
determined by substituting values in equation ___. The same method was used
for determining the fluoride concentration in toothpaste.
Preparation for Standard Addition
A 10-ppm F- solution was prepared by diluting 2.5 mL of a 1000-ppm standard into
a 250-mL volumetric flask. From the prepared solution, 5.0, 10.0, 25.0, 50.0, and
75.0 mL aliquots were transferred to a 100-mL volumetric flask and added with 1
mL TISAB solution and 2mL of the unknown sample (Listerine) then diluted to the
mark. A blank was also prepared by adding 20 mL of the distilled water to a flask.
Then, the prepared standards were each transferred to a 100-mL beaker and
immersed in electrode for 3 minutes until a potential value was stabilized. The
potential (mV) was then recorded.
