BSE IV – BSE BIOLOGICAL SCIENCE MICROBIOLOGY AND PARASITOLOGY

ACTIVITY NO. 1
PREPARATION OF CULTURE MEDIUM

I. OBJECTIVES
At the end of this experiment, the student should be able to:
a. make a culture medium for the growth of the bacteria.

II. MATERIALS
 Personal Protective Equipment (PPE)
 Nutrient agar powder
 Erlenmeyer Flask
 Petri dish
 Spatula
 Stirring rod
 Distilled water
 Autoclave Machine
 Analytical Balance
 Watch glass

III. PROCEDURE
1. Weigh 2.8 g of nutrient agar powder using analytical balance.
2. Add the 2.8 g of nutrient agar powder in 100 mL distilled water placed in
Erlenmeyer flask.
3. Heat the Erlenmeyer flask with nutrient agar powder in a heat bath and stir it
using the stirring rod until it dissolves.
4. Cover all the petri dishes and Erlenmeyer flasks with clean paper. Make sure that
the mouth of the Erlenmeyer flask is covered by a crumpled clean paper.
5. Put the petri dishes and Erlenmeyer flasks in the autoclave tray. Autoclave the
materials for at least 40 minutes in order to become sterile.
6. Light up the alcohol lamp to sterile the workplace.
7. After 40 minutes, release the pressure of the autoclave machine. Then, open the
autoclave machine and get the tray where the materials are placed.
8. Open the Erlenmeyer flask and heat it in the alcohol lamp before transferring the
nutrient agar solution in the petri dish. Make sure that before closing the petri
dish, heat its mouth first.
9. After closing the petri dish, wait for at least 30 minutes in order for the solution to
solidify.
IV. GENERALIZATION
The nutrient agar is very essential in the growth of the bacteria. This provides all
the nutrients required for the bacteria to grow and reproduce. As we all know,
bacteria are very fastidious because they have high nutritional requirement in
order to grow and reproduce by binary fission. Nutrient agar has proteins, sugar,
and fats which sustains the requirements of bacteria in order to survive.
