LAB #6 MACRO METHOD

Give at least 3 sources of technical error in macrohematocrit determination, explain each.

 Blood must be an EDTA-treated. Because EDTA is preferred anticoagulant for hematological test
which is need in hematocrit determination.
 Bubbles in the Wintrobe tube. It is important to fill the Wintrobe tube up to 10 marks without
the presence of bubbles to avoid inaccurate hematocrit readings.
 Read the height of patients packed RBC do not include the buffy coat in the measure for
accurate hematocrit result.

LAB#6 MACRO METHOD

1. I learned that macro method in Hematocrit determination uses a large amount of blood which is
place in EDTA tube. Wintrobe tube was filled up to 10 mark without bubble to prevent
inaccurate result. The centrifuge is calibrated to 2,000-2,300 RPM for 30 minutes to make sure
that the RBC was packed and the hematocrit was read by our eyes in eye level.

LAB #6 MICRO METHOD

1. Micro method for me is the most effective way for hematocrit determination because you
can get result in 5 mins and it also need a small amount of blood. It is easy to read because
of microhematocrit reader.
