MICROSCOPIC EXAMINATION OF URINE SAMPLE:

Microscopic examination on urine ordered


by the physician results may provide
additional information regarding the status
of carbohydrate metabolism, kidney and
liver function and urinary tract infection.
SPECIMEN COLLECTION:

1- Requirements for patient preparation: none


2- Specimen type: first morning is preferred but
random mid-stream urine in a clean container is
acceptable.
3- Storage conditions in room temperature until
tested, store at 2-8°C if greater than 2 hours.
STEPS TO EXAMINE THE URINE SAMPLE UNDER
THE MICROSCOPE:

 1- Receiving the sample.


 2 - Matching the sample with the examination request
paper.
 3 - Shake the cup well until it becomes homogeneous
and contains the components of urine.
 4- Place the tube in the Centrifuge Place another tube in
the corresponding cell with the same amount of urine
from another patient or water to balance.
 5- Set the speed of the device at (1000-1500) rpm for
five minutes.
THE COMPONENTS WE ARE LOOKING FOR BY A
MICROSCOPE IN THE URINE SAMPLE:

A-Salt crystals:
 Some salts are formed in the of alkaline urine,
such as:

 1-Triple phosphate.
2-Amorphous phosphate.
 Some salts are composed in the of acidic urine,
such as:
1-Amorphous urate.
2-Calcium oxalate.

3-Uric acid.
Epithelial cells:
C-CASTS:

1-hyaline
2-granular
3-waxy
4-fatty
 bloody casts
 WBCs casts
Pus cells

Fungi, bacteria and parasites.
