EFFECTIVENESS OF JAVA ACID FRUIT EXTRACT (T. indica L.) ON THE CALCULATION OF LEUKOSIT AMOUNT DIRECT METHOD Supervisor I: Nursidah, SKM., M. Ked Supervisor II: Titi Purnama, S. Si., M. Kes XVIII + 63 + 17 + 9 + 9 Laboratory tests that are often carried out are blood tests, one of which is counting leukocytes. Leukocytes are counted per unit volume of blood by making dilutions of blood using Turkic solutions. Turkic solutions are often unavailable or expired, so research is conducted to look for alternative reagents. The purpose of this study was to determine the effectiveness of variations in the concentration of extracts of tamarind (T. indica L.) on the number of leukocytes. This type of research is experimental research conducted at the STIKES Mandala Waluya Kendari Integrated Clinics Laboratory. The number of samples is as many as 24 counting rooms with manual inspection methods using counting rooms that are examined under a microscope. The data obtained were analyzed by simple linear regression test. The results showed that of the 24 dosage rooms, the average number of leukocytes in the control solution was 9,208 cells / mm3, the concentration of Javanese acid extract was 50% with an average yield of 8,408 cells / mm3, the concentration of Java extract was 70% with the average yield was 6,391 cells / mm3, and Javanese acid extract concentration was 100% with an average of 5,983 cells / mm3. Hypothesis test results show that the sig value is 0,000 <α (0.05), so there is an effective concentration of tamarind fruit extract (T. indica L.) on the direct method of leukocyte count, H1 is accepted and H0 is rejected. Based on the results obtained, the extract of Java tamarind concentration of 50% is the most effective concentration to replace glacial acetic acid in Turk solution. Suggestions for future researchers to use lower concentration variations and pH variations to determine the most effective concentration and pH.
