"Isolation and Identification of Thermophilic Bacteria Phenotypic in Karede Hot
Springs Tourism in Eensumala Village, Bonegunu District, North Buton Regency"

Karede Hot Springs is a hot spring tour located in Eensumala Village District
Bonegunu North Buton Regency. This hot spring is used as a mainstay tourist
destination for the surrounding community. This tour takes the form of a pond
measuring 8 x 10 meters and a depth of ± 2 meters. This study aims to determine the
presence of thermophilic bacteria using culture methods, gram staining, biochemical
tests and phenetic tree construction. This phenetic tree uses two programs, PFE and
MVSP.
The research sample was taken from two points namely surface and base point
with a temperature of 40 ° C. The results obtained were 8 isolates, of which 3 clusters
were formed. Cluster 1 consisted of P1A and D1A isolates with a value of 85.3%,
cluster 2 consisted of P2B and P2C with a similarity value of 82.4%. And cluster 3
consisted of D2A and D2C isolates with a similarity value of 82.4%.
Based on the results of the study it can be concluded that there are
thermophilic bacteria in the Karede hot spring tourism. It is hoped that further
researchers will continue to the molecular stage so that they can specifically identify
thermophilic bacteria in hot springs.
