PART III: PROJECT PROPOSAL (ANONYMOUS)

Study can be proposed under the condition:

- Drug trials are not allowed
- Animal studies are not allowed
- If drugs are used, only approved drugs can be included in the project

1. Project Title: Malassezia composition in healthy scalp of hijab and non-hijab wearer_____________________________

2. Research Subject: Scalp health__________________________________________________________________________

3. Date of start - completion: (01/01/2022) – (12/01/2022)

4. Research category: _____________________________________________________________________________________

5. Summary of the proposed research (Include aims and objectives and rationale for conducting the study, up to 150 words)
Malassezia is a lipophilic yeasts residing on the human skin. Malassezia yeasts are most often found in sebum-rich areas of the skin
such as the trunk, back, face, and scalp. Exogenous factors include heat, humidity, skin covering by cosmetics or clothing, where there
is an increase in sebum, and pH could facilitate Malassezia’s change into mycelial form. The hijab, a veil worn by Muslim women to
cover their heads, is used by most of Indonesian women. Wearing hijab is predicted to change physiology of the scalp since it is
regularly worn. The purpose of this work aimed to describe the composition of Malassezia spp. isolated from the skin of the hijab wearer
differs from that of non-hijab wearer individuals, moreover, we intend to analyze the transepidermal water loss (TEWL), stratum corneum
hydration, and skin pH as a measurement of scalp physiology.

6. Detailed research plan:

a. Design of study (including the total number of cases/samples to be studied and the mode of selection of subjects):

C1 - Internal use
Cross sectional study, observational, descriptive study.
Total sample will be fourty female subjects consisted of 20 hijab wearing and 20 non-hijab wearing subjects with matched age.
b. Equipment and other materials to be used:
Sebumeter, Tewameter, Corneometer, pH measurement, cotton swab
c. Methodology (in detail):
Sample collecting: Sample will be collected from a healthy individual whom wear hijab and non wearing hijab. The sample will be
collected by a cotton swab from 4 designated area and will be transported for culture.
Sebumeter, tewameter, pH measurement and cornemeter will be a non-invasive measurement of scalp physiology.
d. Project schedule (including dates of preliminary and final reports):
November 2021
December 2021
January 2022
e. Techniques to be employed for evaluating the results (including statistical methods)
Malassezia species will be identified using culture and scalp physiology such as TEWL, sebum profile, and skin hydration will be
measured non-invasively with a tewameter, sebumeter, and corneometer, respectively, in four designated area of the scalp. Skin
surface pH will be measured with a pH meter. The paired Student’s t-test will be used with two-tailed P-values and significance
accepted as P < 0.05. Data were presented as mean ± standard deviation.
f. National transfer of biological material to outstation labs (whether required or not):
None

7. Expected outcome and its utility to Indonesia or global dermatology field:

The expected outcome is an added knowledge of Malassezia role in maintaining a healthy scalp in an condition. Morover, by
knowing the physiological baseline in a healthy scalp in Indonesia climate that beneficial for Malassezia growth, practicing a good
care hygiene is mandatory.

8. Present knowledge and relevant bibliography relating to the research topic:

C1 - Internal use
The correlation between Malassezia species and their involvement in skin diseases such as dandruff and seborrheic dermatitis has
been researched. Some studies mention that Malassezia can changed into its mycelial form under some condition, one of them is
usage of clothing that form an occlusion, that can exacerbate scalp disease such as dandruff and seborrheic dermatitis.

9. Links with other projects (Ad-hoc, task force or collaborative; please specify)
