ACTIVITY: CULTIVATION OF BACTERIA

Give examples of the following

A. Selective media
 Selective media contain particular ingredients that inhibit the growth of certain
microbes. An example of a selective medium is MacConkey agar. It contains bile
salts and crystal violet, which interfere with the growth of many gram-positive
bacteria and favor the growth of gram-negative bacteria.
 Eosin methylene blue contains dyes that are toxic for Gram-positive bacteria. ...
 YM (yeast extract, malt extract agar) has a low pH, deterring bacterial growth.
 MacConkey agar is for Gram-negative bacteria.
 Hektoen enteric agar is selective for Gram-negative bacteria.

B. Enriched media
Enrichment media allow the growth of a particular type of microorganism in the
medium. The examples of enriched media are blood agar, chocolate agar, Loeffler's
serum, etc. The examples of enrichment media are Selenite F broth, tetrathionate broth,
alkaline peptone water (APW)

Discuss when to use the following

A. Agar medium
Agar, or agar-agar, is widely used as a culture medium for growing micro-
organisms. The stuff itself is also the product of micro-organisms. Agar, or agar-agar, is
widely used as a culture medium for growing micro-organisms. The stuff itself is also the
product of micro-organisms.

B. Semi solid medium

Semisolid medium has a soft custard-like consistency and is useful for the
cultivation of microaerophilic bacteria or for the determination of bacterial motility.

C. Broth medium
Broth cultures are liquid cultures used to grow bacteria in laboratories. To create
a broth culture, a scientist begins with a sterile liquid growth medium. The medium is
inoculated with bacteria and placed in an incubator at the appropriate temperature
