Work Plan

Duties and Time frame of internship

Knowledge, skills and competences to be acquired:

The trainee will acquire knowledge in the area of cell culturing of mammalian cell lines, plasmid preparation and
the transfection of cells with polymer-plasmid complexes that encapsulate reporter genes. The trainee will learn
to perform cell-based assays (e.g. alamarBlue and MTT cytotoxicity tests), gel permeation chromatography, gel
electrophoresis, hydrogel preparation and will acquire the competency to keep records of the experimental data
and design scientific presentations for group meetings. Other relevant learning outcomes include introduction to
laboratory safety rules and regulations as well as an overview on the current state-of-the-art scientific literature.

Detailed programme of the training period:

Preparation of cell-based hydrogels for wound healing applications

Week 1-15: Establishment of cell cultures (fibroblasts, ADSCs, etc.) to support the further development of cell
encapsulation into hydrogel I wound healing applications Week 4-15: Hydrogel preparation and characterization
of components, secretion of growth factors from cells encapsulated into the hydrogel Week 8-15: Transfection of
cells with reporter genes and preparation of polymer/ plasmid complexes
Deliverable D.1: Hydrogel with encapsulated cells
Deliverable D.2: Genetically modified cells encapsulated in hydrogel

Tasks of the trainee:

• Cell culturing of mammalian cells
• Hydrogel preparation
• Performing cell-based assays (e.g. alamarBlue, MTT cytotoxicity tests)
• Gel Permeation Chromatography (GPC)
• Gel electrophoresis
• Participating in group meetings
• Plasmid preparations
• Reporter gene studies
• Keeping records of experiments, results and maintaining lab books
• Reading relevant scientific literature
• Attendingand
Monitoring of lab safety presentations
evaluation plan:
The trainee will perform his duties/ tasks in the laboratory of the supervisor, Dr. Wenxin Wang, at the Charles
Institute of Dermatology, University College Dublin (Higher Education Institution) where he will have the
opportunity to interact with the mentor and supervisor on a day-to-day basis. The trainee will also take part in
the scientific group meetings where he will give presentations on his work and will receive feedback from the
entire team. In addition, the laboratory manager of the institute will advice on all practical issues associated with
the scientific work in the research group.
Evaluation sheet of sending institution that is filled out at the end of the internship by supervisor at the host
organisation covers:
• Understanding of internship activities and tasks;
• time management&efficiency;
• flexibility;
• communication&teamwork;
• self-organisation;
• focus&perseverance

I confirm that the trainee is not financed by EU money and does not work within an EU project.
