1. Explain an important thing to remember as you turn the high-power objective into
place.

As we turn the high-power objective of a microscope into place, we should


remember that it is retractable and will slid inside if we hit the slide. So, precautions
must be taken to ensure that we did not hit the slide in the process of observing
the specimen.

2. What should you always remember when you use the coarse adjustment?

When we use the course adjustment we should always remember that it


should only be used with the lowest power objective lens. We will only need to use
the fine focus after it is in focus. Using the coarse focus with higher lenses may
result in crashing the lens into the slide.

3. Under what conditions would you adjust the diaphragm?

A microscope's diaphragm, which we're utilizing, needs to be adjusted when


we need to vary the intensity and the size of the cone of light that is reaching the
slide on which we have our specimen.

4. What should you always remember when handling microscope slides?

When handling microscope slides we should always remember not to hold


it in the center because if we do, the slide will become dirty and the specimen we'll
put on it will become dirty as well, and when we look at that specimen with the dirty
slide under the microscope, it won't be able to show us a clear image.

5. What is the purpose of the stage clips?

The purpose of the stage clips is to hold the slide in place. They have a
spring-loaded mechanism that holds the slide's edges in place and locks it in place,
allowing the stage controls to smoothly alter the slide's position.
6. In terms of your eyes, what should you try to learn as you use the microscope?

In terms of our eyes, we should try to learn as we use the microscope to


always keep both eyes open (it takes some practice) to avoid our eyes being
damaged and for those who wear eyeglasses never let the glasses strike the
eyepiece lens to keep the lens of the eyepiece from being damaged as well.

7. What are the two parts used to carry the microscope?

The base and arm are the two parts that were utilized to carry the
microscope. Grab the microscope's arm with one hand and position the other
beneath the base to carry it.

8. What is the purpose of the coverslip?

The main function of the coverslip is to ensure that specimens are


presented in a flat layer of even thickness, this minimizes the need to re-focus the
microscope as different regions of the specimen are viewed. Coverslip also protect
both the specimen and the objective lens of the microscope from damage.
Moisture will ruin the seal around the edge of the lens unless a lens is specially
made to focus through water or oil.

9. What is the objective lens used to locate the specimen and first focus?

The scanning objective lens is the one that is used to locate the specimen
and first focus. Something (sometimes it's only a color) will be shown on this
setting. We can use the course knob to focus: the image may be small at this
magnification, but without this first step, we won't be able to find the image at higher
powers. Move the mechanical stage till our focused image is centered as well.

10. What are the chemicals called that are sometimes used to make the specimens
visible?
Stains or staining reagents are a group of chemicals that are used to make
a specific part of a specimen visible, such as safranin o, crystal violet, and
methylene blue. The chromosomes, the cell wall, and other parts of the specimen
to be investigated are revealed by each stain.

11. What should you do if the high-power objective lens touches or breaks the
coverslip?

We should report to our teacher for inspection if the high-power


objective lens we're using breaks or touches the cover slip. It is important to
be careful when using a high-power objective lens; however, if the lens breaks or
contacts the cover slip, we should immediately notify the teacher, who will assess
the damage and make recommendations for repairs and replacement.
