Microscopic Studies
Microscope usage
Analysis:

1. Set your microscope to a flat surface.

2. Set your microscope's mirror or light, diaphragm and condenser.

3. Set your letter sample between slide and lamel and place on the stage.

4. Engage the 10x objective in the light path. 5. Bring the specimen (letter) in focus.

Examine your letter sample with the smallest objective. Note if the appearance is inverted or not. The letter
placed on the microscope, compare what you see. Note the differences. This will be helpful in the next
microscopic studies in pulverized drugs analysis.

For future studies with pulverized drugs, if you use Chloral Hydrate or Sartur reagents, you have to heat
your preparat gently over a flame to clarify and to remove the air bulbs.

Questions:

1. Describe the mechanical and optical parts of a microscope.

Mechanical parts refer to the parts supporting the microscope. Optical parts refer to the parts that involve
the formation & magnification of image. Mechanical parts include the base (foot), a u-shaped structure
where the microscope stands on. Pillar, a perpendicular projection where the stand lies on the stage. Arm,
this part is a curve-shaped structure that carries the whole microscope. Optical parts typically consists of an
illuminator, condenser, specimen, objective, eyepiece, and detector.
2. If you use a 15x ocular lens and 40x objective how do you calculate the magnification scale for your
microscope?

Magnification is the ability of a microscope to produce an image of an object at a scale larger or smaller than
its actual size. It is indicated that the object is enlarged 15 times and 40 times, thus, the formula is X = ocular
x objective lens power.

x = 15 * 40

= 600x
