EXPERIMENT NO. 1
OPERATION OF THE MICROSCOPE
DATA OBTAINED:
Draw and label the different parts of the microscope. Give the function of each part.

Body Tube- It holds the Ocular lenses and


connects to Objective lenses.
Ocular Lens ( Eye Piece) - It enables you to
see an object through its lens magnifying to
10x. Optional are from 5x-30x.
Revolving Nosepiece - It rotates to choose
another objective lens.
Objectives Lens- This are primary optical
lenses in the microscope for viewing
objects that magnifies to 4x- 100x
Stage Clip- It is to hold the slide to view
the specimen carefully.
Diaphragm- It is where the light passes
through and is able to see the specimen in
place.
Light Source- It is where the light will help
to see the specimen clearly.
Arm- It supports the Body tube.
Stage- It is where the specimen is viewed
in place.
Coarse and Fine Adjustment Knobs- It is to
focus the specimen in the microscope.
Base- It supports the whole component of
the microscope.
LABORATORY GUIDE QUESTIONS:

1. Why is the letter “e” inverted when viewed under the microscope? See Image 1 for reference.
It is because there are two lenses inside the microscope and the placement of the lens is at distance, the visual
rays will bend, making the image as why the letter “e” is inverted.

2. Identify the different type of microscopes and give their uses.


There are two main types of microscope , the Compound Light Microscope and the Electron Microscope.
Compound Light microscope allows light to pass through an image and use two lenses to magnify object while
Electron Microscope uses beams of electrons to form an image that can magnify up to 500000x their actual size.
There are two types of Electron Microscope , the Transmission Electron Microscope (TEM) and Scanning Electron
Microscope (SEM) .

3. Give the advantages and disadvantages in using the electron microscope.


The advantages of an electron microscope is because of the powerful magnification that contributed a lot of
studies and gain more knowledge in the fields of Chemistry but some the disadvantage of it is the cost, use of too
much energy, sensitivity to external magnetic fields and how large it is.

4. Explain how you would view the leaf specimen (See Image 2) in a microscope. Explain how the first view (10x)
was done and then to the second view (40x).

The first view of the leaf specimen in 10x is we can see here the formation of circular lines or different
shapes of the leaf specimen but if we look closer in the second view in 40x, we can see clearly the formation of
stomata in the leaf specimen.
