HEMATOKXYLIN-EOSIN (HE) STAINING PROCEDURE

1. Slide into xylen solution (Bottle no 1,2,3,4) 3 to 5 minute each
2. Slide into alcohol solution (Bottle no 5,6,7) 3 to 5 minute each
3. Wash Slide in tap water (Bottle no 8) 5 minutes
4. Slide into hematoxylin solution (Bottle no 9) 5 minutes
5. Wash Slide in tap water (Bottle no 10) 3 minutes
6. Slide into eosin solution (Bottle no 11 ) 5 to 10 dip
7. Slide into alcohol solution (Bottle no 12,13,14,15) 2 to 3 dip
8. Slide into xylen solution (Bottle no 16,17,18,19) 1 to 2 minutes
9. Mounting

The above is hematoxylin-eosin staining procedure.

It requires 19 bottle consisting of different predetermined liquid, they are 8 bottles of xylen, 7
bottle of alcohol, 2 bottle of water, 1 bottle of hematoxylin, 1 bottle of Eosin,and 1 mounting.
First immerse the slide into xylen in bottle 1,2,3,4 for 3-5 minute each. Then immerse the slide
into alcohol from bottle 5,6,7 for 3-5 each. After that rinse with running tap water for 5 minutes
in bottle 8. Next step, immerse the slide into hematoxylin solution for 5 minutes in bottle 9,then
wash again in dip water for 3 minutes in bottle 10. Then dip the slides into eosin solution for 5-
10 times in bottle 11, after that immerse the slides into alcohol in bottle 12,13,14,15, then into
xylen solution in bottle 16,17,18,19. And the last step is mounting. Mounting is to cover the slide
using glass and glue cover.
