Exercise No. 1: THE COMPOUND MICROSCOPE AND OTHER

INSTRUMENTS IN MICROBIOLOGY

Name: Fretzson G Cotdi. Date: 24/01/2022

Year and Section: BSND 1B Group Number:

Instructor/ Professor: Jennifer Paltiyan

1. Carefully study the different parts of a microscope. Based on the description and
other references, label the parts of a compound microscope in your work sheet.

1. Revolving nosepiece 5. Diaphragm 9. Arm

2. objectives lenses 6. Light source 10.Coarse adjustment
3. Stage clips 7. Eyepiece 11. Fine adjustment
4. Stage 8. Body tube 12. Base
2. Let us assume that you will be using a compound light microscope for your activity.
Upon checking, the microscope which you obtained from the preparatory room has
the following specifications:
* Ocular lens magnification= 10x
* Scanner magnification= 4x
* Low power objective magnification= 10x
* High power objective magnification= 40x
* Oil immersion objective magnification= 100x

Given this information, complete the table below: (12 points)

INITIAL FINAL
OBJECTIVES TOTAL MAGNIFICATION
MAGNIFICATION MAGNIFICATION
Scanner 4x 10x 40x
Lower Power 10x 10x 100x
Higher
40x 10x 400x
Power
OilImmersion 100x 10x 1000x

3. Identify the commonly used instruments in the Microbiology Laboratory. Indicate

their uses.
Name of instrument Uses

a Wire loop Wire loop is used to smear, streak, or take an

inoculum from, a culture of microorganisms.

b Incubator incubators are also used to increase the

growth rate of organisms, having a prolonged
growth rate in the natural environment.

c Culture media Some culture media are considered general all

purpose media and support growth of a large
variety of organisms.

d Microincinerator Microincinerator This product uses infrared

heat in a ceramic tube to protect lab
technicians from dangerous gases, flames and
splashes.

e Bunsen burner It is a gas burner used in laboratory and it is

used for experimenting it produces a single
open gas flame, and is used for heating,
sterilization, and combustion.

f Pipettes Uses to transport a measured volume of liquid

g Culture tube It is often used in biology for handling and

culturing all kinds of live organisms, such as
molds, bacteria and in medicine and forensics
to store samples of blood or other fluids.

Question for research

1. What are the important uses of a microscope in Microbiology and Parasitology?

(10 points)

- the important uses of microscope in microbiology and Parasitology is that without

using microscope in microbiology how can we see the small things like bacteria or
little species that living in the object and microscope is also an important diagnostic
tool in parasitology. Electron microscopes are also useful but more as a research tool
than for diagnostics Parasites, their eggs or larval stages can be identified in blood,
faecal or tissue samples, often with great accuracy.
2. What is the function of the iris diaphragm? To what part of the human eye would
you compare it? (10 points)

I think The function of the iris in the eye is to control the amount of light that reaches the
retina. It does this by changing the size of the pupil
