Topic: Effect of temperature on rate of an enzyme catalyzed reaction.
Introduction
Aim: To determine the effect of temperature on the rate of an Enzyme catalyzed reaction using
catalayse solution and Hydrogen Peroxide.
Materials/ Apparatus: Thermometer, beaker, boiling tubes, Styrofoam cup, wire gauze, bunsen
burner, 30% catalayse solution, 6% Hydrogen Peroxide, tripod stand, ruler, tap water, ice,
measuring cylinder
Method:

1. Set up water baths at temperature ranges 0-5 , 10-15 , 20-25 , 30-35 , 40-45 ,

50-55 , 60-65 , 70-75 .


2. Pour 1 cm3 of liver (catalase) solution into a boiling tube and 10 cm3 of 6% Hydrogen
Peroxide into another boiling tube.
3. Place both test tubes into a water bath and allow to remain until the temperature of the
catalase solution and H2O2 is the same as that of the water bath. Take temperature
readings periodically and maintain a strict control of the temperature a of the respective
water bath.
4. When the temperature of both solutions is equal to that of the water bath, pour the
hydrogen peroxide solution onto the liver solution while the test tube with liver solution
remains in the water bath. [Do not raise it out of the water bath]
5. After one minute, measure the height of the foam then take another reading 2 minute
following the first reading. Allow another 2 minutes to lapse and take a third reading.
6. Use the results to plot a graph showing effect of temperature on rate of decomposition.
