1. TOPIC: Microscopy and Cells

2. OBJECTIVES:

To describe the parts and functions of the compound light and dissecting microscope.
To state the steps in proper order for bringing the cell’s image into focus with the compound light
microscope.
To calculate the total magnification and the estimated size of the Cell’s image.
To identify the differences between cells and classify them.

Research Questions: What is the estimated size of cells?

Hypothesis: The size of the cells is very small to be seen by naked eye, their size can vary from
one to a hundred micrometers

3. THEORETICAL FRAMEWORK:

A microscope slide is a thin sheet of glass used to hold objects for examination under a
microscope. A standard microscope slide is 75 x 25 mm and 1.2 mm thick. A range of other
sizes is available for various special purposes. A simple light microscope manipulates how
light enters the eye using a convex lens, where both sides of the lens are curved outwards.
When light reflects off of an object being viewed under the microscope and passes through
the lens, it bends towards the eye. This makes the object look bigger than it actually is. A
light microscope can be used to determine the actual cell size. Using a light (optical)
microscope, you can easily determine the cell size.

4. MATERIALS:

Microscope:(PARTS)
Objective lenses (4x, 10x, 40x ,100x)
Immersion oil
Bacteria, plant and animal cell samples

5. PROCEDURE:
✓ Choose Bacteria Gram Stain Mix

✓ Focus the specimen in 4x

✓ See the specimen in 10x, 40x, 100x

✓ Repeat the same process using Plant cells slide and one more time using Human
Blood cells

6. SAFETY PERSONAL AND ENVIROMENTAL MEASURES:

The first suggestion for this experiment is that if you're going to the lab, you should wear a
gown that hasn't been used outside of the lab, tie it tightly, and cover your hands with
gloves. Long hair should also be pulled back.

8. DISCUSSION:

The cell size depends on the type of cell, and each one of the cells has different properties
and parts. Most of the cells can be easily seen with the 100x objective lens, but you need
the immersion oil to use that objective lens.

9. CONCLUSIONS:

What we can conclude from this experiment is that the cell size can vary from their type, this
meaning that the cell will vary in every other aspect too, most of the time they do.
