DETERMINATION OF SGO-T (AST)

METHOD: Frankel Reitman

PRINCIPLE: The transaminase are enzymes that catalyzes the intra-conversion of keto acids and
amino acids in the process of which the amino acids group is transferred from one molecule to another.
This results in the disappearance of one amino acid and the formation of a new one. The blood serum is
added to a buffer solution of alpha-ketoglutaric acid and aspartic or alanine and resulting oxaloacetic or
pyruvic acid formed after reaction with dinitrophenylhydrazine is measured at 490 -550nm.

4. Aspirate 100ul of
unhemolyzed Test
Sera/Controls in cuvette.

5. Place 250ul of SGO-T


Substrate to 50ul of
unhemolyzed Test
Sera/Controls in
separate test tubes.
6. Incubate for 30 MINUTES at
37-C.

7. Add 250ul of Color Developer


to each tube, mix and stand for
FIVE MINUTES at room
temperature.

8. Add 2000ul of 0.4N Sodium


Hydroxide, mix and read against
Water Blank after FIVE MINUTES.

Interpretation: The normal range for determing SGO-T(AST) is 0-40 U/L.


Therefore, Ms. Winnie Navarro, patient female adult with 9.23
U/L has a normal value for SGO-T (AST) and doesn’t have any liver
damage or disease prevalence.
