DETERMINATION OF SPECIFIC AGGLUTINATION TITRE OF COMMERCIAL ANTISERA

Serial dilution is a series of simple dilution in which the concentration of a substance


decreases by the same quantity in each successive step and is used in the determination of antibody
titre.

The sample was initially determined, labelled


type A and type B. Blood collection was done
via venipuncture.

Multiple test tubes were utilized, containing


NSS. Each was labelled from the dilution ratio
1:1 through 1:152. A red cell suspension was
prepared.

The Anti-A, Anti-B and 5% cell suspension were


mixed in each tube and were consecutively
transferred one tube to the other with each
achieving 0.2 mL volume of liquid.

After centrifugation, the following appearances


of the mixtures were obtained.

Results:

The titre of the blood type A falls unto the ratio


1:4, meaning that one part serum to 4 parts
saline solution (dilutant) results in no
antibodies remaining detectable in the next
tubes.

The titre of the blood type B falls unto the ratio


1:16, meaning that one part serum to 16 parts
saline solution (dilutant) results in no
antibodies remaining detectable in the next
tubes.
