BROAD SPECTRUM COMPATIBILITY TEST
PRINCIPLE: Broad-spectrum compatibility test in blood banking is to determine the compatibility
between the donor's blood and the recipient's blood before a blood transfusion takes place. This
test is commonly known as a crossmatch. It involves mixing a sample of the donor's blood with a
sample of the recipient's blood and observing the reaction between the two. The purpose is to
identify any potential incompatibilities that could lead to an adverse immune response in the
recipient if incompatible blood is transfused.
PROCEDURE:

A. Collection of Specimen

1. Collect 2 same blood type samples for the


Donor and Patient specimen using Syringe
Venipuncture Procedure

B. Protein/ Albumin/ Room temperature


phase (immediate saline spin)

1. Label 4 wasserman tubes as M, MA, N and


NA

2.
In Tube M, contains 2 drops of patient's serum
and 1 drop of 2-5% RCS of donor's cells.
In Tube MA, contains 2 drops of patient's
serum, 1 drop of 2-5% RCS of donor's cells and
2 drops of BSA.
In Tube N, contains 2 drops of donor's serum
and 1 drop of 2-5% RCS of patient's cells.

In Tube NA, contains 2 drops of donor's serum,


1 drop of 2-5% RCS of patient's cells and 2
drops of BSA.

3. After mixing, centrifuge the tubes for 15


seconds at 3400 rpm.

4. Gently dislodge the cell button and observe


agglutination and if no agglutination MA and
NA tubess will proceed to THERMO PHASE.

C. Thermo/ Incubation Phase (37⁰C)

1. Incubate MA and NA tubes for 30 minutes at


37⁰C

2. Centrifuge for 15 seconds at 3400 rpm

3. Gently dislodge and if no agglutination is


observed in tubes MA or NA proceed to AHG
phase.

Based from the results, there is no


agglutination or hemolysis observed.
Interpretation:
Therefore the Donor and Patient is
COMPATIBLE for Blood transfusion.
