Animal Protein Classification
◦ Sarcoplasmic protein: water soluble protein, components of the
intracellular fluid
◦ Myofibrillar protein: salt soluble protein, main component of meat
protein
◦ Stroma protein: connective tissue protein, the residue after
extraction of the sarcoplasmic and myofibrillar protein

Sarcoplasmic protein
◦ Water soluble component
◦ Intracellular fluid liquid contains
different types of enzyme,
pigments, parvalbumins, antifreeze
proteins, marine toxins and NPN
◦ The content of sarcoplasmic protein
is generally higher in pelagic fish
and lower in demersal fish
◦ When separated by
electrophoresis, sarcoplasmic
proteins can be used to fingerprint
or identify fish species

Myofibrillar proteins
◦ Salt soluble component
◦ Myofibrillar proteins constitute more
than 60% of the total muscle
proteins and include mainly myosin
and actin
◦ Contribute to the muscle shortening
and gelation properties
◦ Fish muscle myofibrillar proteins are
thermally less stable than those
mammalian muscles

Myosin
◦ The myosin from skeletal muscle of fish
are similar in structure to mammalian
and avian
◦ Myosin is the principals myofibrillar
proteins, making up over 40% of the
total and it is located in the thick
filament
◦ The molecular weight of myosin is 200
kDa
◦ In the formation of the network during
setting, a major role is played by the
aggregation of the myosin through
different protein-protein interactions

Actin
◦ Actin is the myofibrillar protein of
second highest contraction and it is
found in the thin filaments along
with tropomyosin and troponin
complex
◦ In the fish muscle, actin constitute
about 20% of the total myofibrillar
proteins
◦ The molecular weight of myosin is
45 kDa
◦ Actin itself, contribute less or no
gelling ability

Stroma protein
◦ Stroma is the protein which forms
connective tissue, it cannot be
extracted by water, acid, alkaline
and salt solution
◦ A similar composition found in
mammalian, but fish muscle
contains less stroma than
mammalian
◦ Collagen and elastin
◦ Marine collagens have a higher
absorption capability (up to 1.5
times more efficiently into the body)

Marine collagen
◦ The most abundant type of collagen in
the body, type I, can be found in the
bones, skin, tendons, and organs.
◦ Type II can be found in cartilage, and
type III can be found in reticular fibers,
blood, and skin.
◦ Collagen type III can be found in the skin,
vessel walls, and reticular fibers of the
lungs, liver, and spleen
◦ Type I and V, have been identified in the
intramuscular connective tissue of fish
muscles
◦ Type V is more labile to enzymatic
degradation during cold storage and
causes postmortem softening
