Process

1) Set software parameters; Create a long cycle time say 100secs (x2, normal cycle time ~
55sec), set sample time to 5secs and inject time to 50mS.

2) Set columns Temperature and Pressure; Method = working on guess work and correction,
but commonly a good start point is at 90degrees and 20PSI for both and work up.

3) Adjust Separation/Spacing and Max peak values; Using Temperature.

Temperature = Separation + Peak Max

4) Adjust Elution Time (cycle time); Using Pressure.

5) Adjust Peak Height Further; Using Sample time.

6) Adjust Sensitivity; Using Inject time, warning too high value will lead to peaks going
outside A/D range and be cut. Too low a value will mean peaks unstable.

7) Optional Adjustment; Injector and Inlet temperature, usually 100 to vaporise water but can
be adjusted for performance. But do not go below ambient temperature in the box or cromat
will never sample (as it can never reach desired temperature.
8) Adjust clarity of later samples; Using backflush (flushing out leftover gases from last
sample). Best to start with a high time and work down to the end peak just disappears.

9) Calibrate Peak Ranges and Identification; Click “View” on top bar and tick “ show cal
points” and define peak margins by using the calibs tab and setting the start and finish point
for each peak (one at a time). Check and correct until each sample puts peaks in the correct
range automatically. (use sample on/off buttons to pause, on the control cab).

10) Check if program detects peak points correctly; If not adjust the ranges again. Note some
peaks are so close they the ranges you selected may over lap, if so adjust manually the
figures and give at least a 3 point spacing for safety.

11) Let Auto Calibrate fine Tune the Settings; Go to “Control” tab and click on the “Chrom”
button. Check the results. Chromat is then calibrated.
