Practical # 01: Separation of components of a mixture through TLC plate

Procedure:
1. First dissolve all the samples (mixture) you want to run (solids and liquids) in small
beaker using a volatile solvent in which they are soluble.
2. Obtain a TLC chamber with lid. An inexpensive chamber can be made using a beaker and
watch glass.
3. Add a portion of a prepared solvent for chromatography to the chamber or beaker.
4. Take a TLC plate, Use a rule to lightly draw a straight line with a pencil roughly 2cm
from the bottom. Label the areas with pencil where you plan to place the samples.
5. Using a capillary tube spot the sample (mixture) on TLC plate at different positions.
6. Place the TLC plate in the chamber for the plate development. The liquid level must be
below the pencil line where the samples are spotted.
7. After elution, remove the TLC plate from chamber and locate the different spots of the
mixture components.
8. If the components are colourless , visualize it with the help of visualizing agents.
Practical # 02 : separation of components of a mixture through radial
Chromatography
Procedure:

1. Take a circular filter paper and make a small hole right in the center of it.
2. Make some dots of ink mixture around the center of circular filter paper with the help of
syringe
3. Now prepare a wick of cotton or thread and place it in the center of circular filter paper.
4. Pour some water into the bottom of the petri dish or a beaker and position the filter
paper so that the wick is in the Center of the petri dish or beaker.
5. Wait for several minutes to get different colors bands. Do not let the water or colors
touch the Edge of the paper.
6. Observe the separated colors bands and record it.
