Microbiological Media
The ingredients of the media used in these exercises are in grams per liter of distilled water unless
otherwise specified. Sterilization is accomplished by autoclaving at 15 lb pressure for 15 minutes
unless otherwise specified. Most of the media are commercially available in powdered form, with
specific instructions for their preparation and sterilization.

L-arginine monohydrochloride or
L-ornithine dihydrochloride 10.0 g/l
Salt medium—Halobacterium
Sodium chloride 250.0
Magnesium sulfate • 7 H2O 10.0
Potassium chloride 5.0
Calcium chloride • 6 H2O 0.2
Yeast extract 10.0
Tryptone 2.5
Agar 20.0
Note: The quantities given are for preparation of
1 liter final volume of the medium. In preparation,
make up two solutions, one involving the yeast
extract and tryptone and the other the salts. Adjust
the pH of the nutrient solution to 7. Sterilize
separately. Mix and dispense aseptically.
Urea broth
Urea broth concentrate (filter-
sterilized solution) 10.0 ml
Sterile distilled water 90.0 ml
Note: Aseptically add the urea broth concentrate
to the sterilized and cooled distilled water. Under
aseptic conditions, dispense 3-ml amounts into
sterile tubes.
