Parasite observation

using microscope

Introduction
Some parasites are very small and can only be seen with a microscope
- Using a microscope, parasites are easily observed so that they can be
identified
- Examination of parasites under microscope can be carried out directly
with a wet preparation or by means of a fixed examination with
permanent mount
- In this practicum will show the cyst of Entamoeba coli and
trophozoite stage of Plasmodium vivax belonging to the protozoan
parasite and the egg of Ascaris lumbricoides belonging to helminth parasite
Introduction
- Under microscope, the cyst of Entamoeba coli, the trophozoite
Plasmodium vivax and eggs of Ascaris lumbricoides can be easily
identified by looking the morphological characteristics and their
appendages
- Examination of direct wet preparations can be seen with 100 or 400
magnification while examination of permanent mount can be seen
with 1000 magnification with emersion oil
Objectives
- Recognize cyst morphology of Entamboeba coli microscopically in wet
and permanent mount
- Recognize morphology of trophozoite Plasmodium vivax
microscopically in permanent mount
- Recognize Egg morphology of Ascaris lumbricoides microscopically in
wet mount
Tasks
1. Observe Cyst Entamoeba coli and appendages in wet preparation
(Iodine stain) using microscope and draw the parasite
2. Observe Cyst Entamoeba coli and appendages in permanent stain
(Trichrome Stain) using microscope and draw the parasite
3. Observe Egg of Ascaris lumbricoides and appendages in wet
preparation (iodine stain) using microscope and draw the parasite
4. Observe trophozoite stage of Plasmodium vivax and appendages in
permanent stain (giemsa stain) using microscope and draw the
parasite

Characteristics:
- Size: 15 – 22 µm
- Shape: usually spherical
Sometimes oval
- Number of nuclei: 1- 8, at least 5
nuclei are visible in the shown

Characteristics:
- Size: range, 45-70 µm x 35-45 µm
- Shape : oval
- Egg Wall: thick (outer layer of
albuminoid, inner layer of hyaline)
- Contents: embryos that have not
cleaved
