EXPERIMENT 1: PIPETTING DISCUSSION

Pipetting is a technique used in clinical chemistry which involves transferring of aliquot of liquid
sample from one container to another. Pipettes classified
1. according to designs are:
-to contain
-to deliver
2. according to drainage characteristics:
-blowout
-self-draining
3. according to type:
-transfer pipette (volumetric and Ostwald-Folin pipettes)
-measuring pipette (Mohr and Serological pipettes, for less than 1.0 Ml-MICROPIPETTE)

AUTOMATIC PIPETTE- permit rapid repetitive measurement and delivery of precise volumes
DISPENSER PIPETTE- considered as automatic pipettes that can obtain fluid from a common
reservoir and dispense It repeatedly.

The automatic pipette operates via a stainless steel piston that displaces air, creating a
vacuum that will draw liquid samples into the disposable plastic tips. Using the thumb, the
plunger is depressed to create a vacuum, allowing liquid sample to draw up as it is slowly
released. The plunger is then depressed again to deliver the aspirated liquid to the receiving
vessel.
When manipulating the PLUNGER, two points of resistance are met;
The FIRST STOP is the point when the piston reached the calibrated volume on the dial.
The SECOND STOP is used to fully expel the aspirated volume into receiving vessel
(equivalent to blowing out a glass pipette).
The VOLUME ADJUSTMENT KNOB includes a micrometer to dial in the desired
volume to be aspirated that us displayed in a side window called the volume indicator. The
polypropylene disposable tip fits snugly at the tip of the plastic shaft. The tips may be safely
removed from the pipette using the tip ejector button.
