EXPERIMENT OF BEET ROOTS

Effect of different Temperature

TEST OF PERMEABILITY
 Aim : to study the Effect of different temperature
degree on Cell Membrane permeability

 Material :
1- cork-borer used for cutting of beet roots .
2- test tubes and racks.

Let's start 3- Distilled water .


4-Cutter and Ruler.

5- water Bath .
6- Spectrophotometer - Colorimeter
1- By using Cork-borer we cut Beet root into cylinder shape .
2- By using knife and Ruler cut these cylinders to the same
discs that have the same color , shape and size.

3- Then by using running water wash these discs well.


4- Then Bring test tubes and labelled them according to
condition used " frozen – Room Temp –40°C- 80°C "
Procedures 5-then put 3 ml of distilled water in each tube and put one
disc from beet root discs in each tube .
6-leave them in water bath for 20 minutes after adjusting
condition as you need .
7- write your observation .
8- you can use spectrophotometer if it is available to
estimation the concentration of the color .
Observation
As we increase the temperature
the color increase.
Discussion

1- washing of discs after cutting to remove any colors


that adhere to the surface as a result of cutting.

2- we use a ruler and cutter to have the same size of


discs to avoid any errors .

3- we must take the same discs that have the same size ,
shape and color to avoid errors .
Comment

 Beetroot contain a pigment called


Anthocyanin that give characteristic purple
color to it .
 This pigment found in Vacuoles .
 Increasing of temperature Cause protein to
be denature so it's permeability
increase causing this pigment to
escape into water solution .
As we mention we

Mechanism of Spectrophotometer
Difference  Transmittance: it's the amount of light that “successfully”
passes through the substance and comes out the other
between side.

 Absorbance :is the flip-side of transmittance and states
and how much of the light the sample absorbed.

How can we use this device in our Experiment

1- we must have this device .

2- adjust wavelength of device

3-we use a cuvette of distilled water as a blank .

4- after zero put each solution that in test tube


in cuvette an measure absorbance of light.

5- draw a curve for the result
