Microneedling device :
The microneedling device used in this study
consists of a handpiece , safety needle
cartridge and a needle depth gauge that
allows the user to control the depth incision
from 0 to 1.5mm . The control unit allows
to adjust the frequency of the needle stroke
from 100 to 150hz . The safety needle
cartridge contains six sterile , single use
stainless steel microneedles.
2. Stimuli- Responsive Charge reversal
Nano Drug Delivery System :
With the reference of this article, The
stimuli responsive charge nano drug
delivery system(CRNDDS) for the
treatment of tumors has attacted much
attention as an effective drug delivery
system . The change reversal
nanocarriers are either uncharged or
negatively charged under normal
physiological environment, however
they acquire positive charged in tumor
tissues microenvironment . The process
of charge reversal from negative to
positive results in not quick drug release
in acidic environment but also enhance
the cellular uptake of drug.
The need of stimuli responsive
components in microfluidic system has led
to the development of hydrogel-based
patterned microstructures . The most
commonly practiced means for fabricating
microplatterned hydrogels is based on in
situphotopolymerization using 365nm UV
light in a liquid medium .
3.Thermosensitive hydrogels as
sustained drug delivery system for
CLTA-4 Checkpoint blocking antibodies:
With the reference of this article,
Thermosensitive hydrogels are very
important biomaterials used in drug
delivery systems(DDS) which gained
increasing attention of researches.
Thermosensitive hydrogels have great
potential in various applications such as
drug delivery, cell encapsulation , tissue
engineering etc. .
The potential of CTLA(cytotoxic
Tlmphocyte-associated protein
4)blockade to confer durable response
rate in cancer patients has become
increasingly evident [1,2].CTLA-4belongs
to the category inhibitory checkpoints
molecules, which can be exploited for
generating anti-tumor immune
responses[3,4].It is expressed profoundly
on activated T-helper 1 cells(TH1)and
cytotoxic T-Lymphocytes (CTLs).Upon
binding the antigen presenting cell (APC)
co-stimulatory molecules CD80 and
CD86, CTLA-4 transmits inhibitory
signals, which process leads to
cessations of T Cell activation and
proliferation[5]. As such, blocking the CTLA-
4 receptors and its associated immune
pathways might unleash anti-tumor
immunity, thereby keeping tumor growth in
check. Phase II/III trials indeed
demonstrated robust therapeutic effects. T
Thermosensitive poloxamer 407(P407)
hydrogels were evaluated as slow release
system for optimizing CTLA-4 therapy . Slow
release reduces systemic antibody levels
and potentially mitigates the side effects of
CTLA-4 therapy . The 25% P407 hydrogel is
injectable at room temperature.
4. Size Tunable Strategies for a Tumor
Targetted Drug Delivery System :
With the reference of this article,
Nanoparticles have been widely used in
tumor Targetted delivery while the
antitumor effects are not always
satisfactory due to the limited penetration
and retention . As we all know there
paradox that nanoparticles with large size
tend to distribute around tumor blood
vessels rather than penetrate into tumor
parenchyma , while smaller sizes can
penetrate deeply but with poor tumor
retention. In recent days an intelligent size
tunable strategy provided a solution to
determine the size problem of
nanoparticles and exhibited good
application prospects. Apart from the
applications in oncotherapy size tunable
strategies also have a great prospect in the
diagnosis and real time bioimaging fields
which are also introduced in the review .
Finally the potential challenges for
application and future direction are
thoroughly discussed providing guidances
for further clinical information.
