Materials:
Microscope
Prepared slide (Onion root tip)
Lab Paper
Procedure:
1. Set up a compound light microscope and turn on the
2. Place a slide containing a stained preparation of the onion root tip.
3. Locate the meristematic zone, which is just above the root cap at the very end of the tip.
4. Focus in on low power and then switch to medium or high power. Below find micrographs of
the four stages of mitosis. Use them to help you identify the stages on the microscope slide.
5. Now count the number of cells found in each stage of mitosis and place the data in the chart
below.
6. Determine the percentage of time each cell will spend in each stage of mitosis. Divide the
number of each cell by the total number of cells and multiply by 100 to determine the
percentage. Place these values in the chart.
Stage of Percent of time in each
Number of Cells

Metaphase ____________________________________________ _____________%
Anaphase ____________________________________________ _____________%
Telophase ____________________________________________ _____________%

7. Graph the data (line graph) you have just collected and then answer the questions that follow.
Graph cell stage on x-axis (independent axis) and % of time in each stage on y-axis
(dependent axis)

Analysis & Conclusions:
2. Of the four stages of mitosis, which one takes the most time to complete?
Interphase-
Prophase-
Metaphase-
Anaphase-
Telophase-
4. What evidence shows that mitosis is a continuous process, not a series of separate
events?
5. The onion plant began as a single cell. That cell had X number of chromosomes. (The
exact number does not matter, we will just call that number “X”.) How many chromosomes
are in each of the cells that you observed? (Give the answer in terms of X.) How do you
know?
6. If this onion would reproduce sexually, it would need to produce sperm and/or eggs
by the process of meiosis. After meiosis, how many chromosomes would be in each
sex cell (in terms of X)?
7. If this onion would complete the process of sexual reproduction (fertilizing an egg cell),
how many chromosomes would be in the zygotes that are produced (in terms of X)?
8. What are two reasons why cells divide rather than continue to grow indefinitely?
a.
b.
Events of the Cell Cycle
9. Interphase is divided into what three phases?
a.
b.
c.
Mitosis
12. What are the four phases of mitosis?
a.
b.
c.
d.
Match the description of the event with the phase of mitosis it is in. Each phase may be used
more than once.
Event Phase
_____14. The chromosomes move until they form two groups near the
poles of the spindle. a. Prophase
_____15. The chromosomes become visible.
b. Metaphase
_____16. A nuclear envelope re-forms around each cluster of
chromosomes. c. Anaphase
_____17. The centrioles take up positions on opposite sides of the
nucleus. d. Telophase
20. What is cytokinesis and how does it vary between plant and animal cells?
