INTRODUCTION
Potato crops are susceptible to infection with different viruses, which can result in a reduction of yield and
quality of the tubers. Generally under in vivo conditions infection can be detected by the presence of
symptoms in the leaves, tubers, and plant development. However, sometimes the symptoms do not
manifest themselves due to factors such as the concentration of virus in the plant, the climate or
interactive reactions between the virus and the plant.
To determine whether a plant is infected with a virus, serological (eg. ELISA NASH) or molecular (eg.
PCR) methods are used. In addition, the detection of symptoms in indicator plants is also a method that
can be used. At CIP, a set of 11 indicator plants is routinely used for detecting potato viruses. This set of
indicator plants is known to include at least one host susceptible to each of the mechanically transmitted
viruses in potato. By analogy, any other unknown virus that might be present in the potato under indexing
will stand a high probability of being detected through this procedure.
SCOPE
Some plant species react with very visible and characteristic symptoms when infected with a virus. These
plants can be used as an indicator of virus infection. Some plant species react very visible and
characteristic symptoms when infected with plant viruses, including the potato crop and are as follows:
Gomphrena globosa very susceptible to (PVX) showing symptoms at 5 days after inoculation and from 10
to 15 days in the following plants as Nicotiana glutinosa, Nicotiana benthamiana, Nicotiana debneyii,
Datura stramonium, Nicotiana tabacum "White burley" Chenopodium quinoa, Chenopodium murale,
Physalis floridana and Lycopersicon esculentum. These plants can be used as indicator plants virus
infection.
SAFETY
Disinfect the greenhouse 1 week before and after working a new badge of materials (see point
3.3).
Use laboratory coat.
Wash hands before and after enter to the greenhouse
Indicator plants should be grown in as near optimal condition as possible to stimulate rapid and
luxuriant growth.
Good control of insect vector of virus into the greenhouse (eg. White fly and afids).
Use masks and gloves during the inoculation process.

1.1.2 Propagate in vitro plantlets in a No. 16 tube to evaluate each accession, according to the Propagated
in vitro protocol (OP-056) .The multiplication is performed from the parent plant.
1.2.1. In the greenhouse, place 60 jiffies # 7 in a 24 x 31 x 6 cm plastic tray, hydrated with 4 liters of bencil-
dimetilalquil-amonio (Dimanin ®) solution to 0.5 ml / l. Let soak until they hydrate and make a small hole, the
approximate diameter of a pencil, in the center top of each jiffy.
1.2.2. With the help of two forceps, take out a plantlet from each test tube and remove the culture medium
adhered to the roots. Be very careful not to damage the roots.
1.2.3. Transplant the plantlet in the jiffy and identify it with the corresponding label stuck to the stake.
1.2.4. Sterilize forceps in the flame after each transplant.
1.2.5. Complete transplanting in the tray and add a little distilled water to each plantlet to prevent
dehydration.
1.2.6. Cover the tray with a perforated plastic bag over the middle, fix it with adhesive tape to the tray.
1.2.7. Remove the plastic after 2-3 days.
1.2.8. Place the tray on a table in the greenhouse.
2.1.1.Seed from indicator plants are germinated in clean 24 x 31 x 6 cm plastic trays with six holes in the
base to facilitate water drainage. The trays are previously disinfected with bleach 3%.
2.1.2. Fill two thirds of the tray with soil for the seed bed (Appendix 2). Make 10 rows, 0.5 cm deep,
separated by 3-4 cm from each other.
2.1.3. Use one tray for every two indicator plant species, and position 200 (0.2 g/species)seeds per row.
According to the days necessary for germinating and ideal age or size of the plants ready for inoculation

(see Table 1), start sowing of the indicator species in the 'order of planting' as indicated in Table 1. For
example: Order 1 for Gomphrena globosa; Order 2 for Nicotiana debneyii, N. glutinosa, etc.; Order 3 for
Nicotiana benthamiana; Order 4 for Chenopodium murale, C. quinoa, etc.
2.1.4. Incubate the trays containing the seeds at a temperature of 24-26 ° C until the emergence of the
seedlings. Water regularly to keep the soil in trays wet, with the help of a sprinkler. The time of seed
germination is 4-12 days (Table 1).
2.2.1 When the seeds have grown 2 – 3 true leaves (approximately 4 – 12 days after the sowing), remove
each plant individually of the seed bed and transfer to a pot of 3.5” of diameter. Fill the pots with steril soil
(until the edge), the pots are previously washed and disinfected with bleach 3% (Appendix 1).
2.2.2 Sow the plants in the pots and add more soil. Water immediately with tap water to moist the soil and
then regularly to maintain the plants viable. A week before the inoculation the plants are removed from the
healthy plants production area.

Fumigation is done applying fungicide (benomyl 2g/L, FARMATHE @), and a mixture of pesticides
(imidaclopid 1.5 ml/L, CONFIDOR @; abamectina 2ml/L, VERTIMEC @). Set a yellow trap for insects every
two tables.
3.3. A week before the test, sort 15 ranges per table. Place the indicator plants in a row according to the
order established in Table 1.
3.4. Identify each row of indicator plants with a white sign indicating the name of the species.
3.5. The indicator plants will be inoculated according to the indicated age of the plant or number of leaves
per plant as indicated in Table1
3.1. Sampling
3.1.1 Print a bar-coded label for each accession to be evaluated and stick on a plastic bag (10x14 cm).
3.1.2. Put the labeled plastic bag next each corresponding pot.
3.1.3 With the help of the plastic bag, take a portion of the basal, middle and apical leaf of each accession
to be evaluated. Avoid touching the plant with your hands. Each sample should weigh between 0.5 - 1.0 g.
3.3. Inoculation
3.3.1. Only when the indicator plants have been inspected and found ready for inoculation the bar-coded
label is printed for each accession When printed stick the label on a plastic stake.
3.3.2. Put on latex gloves before inoculation.
3.3.3. Dust carborundum powder (600 mesh) onto leaves of indicator plants.
3.3.4. Place each sample bag containing the extraction sap in front of each host range set, placing the stake
with the accession identification in the first indicator plant from each host range, respectively.
3.3.5. Lightly hold the leaf of the indicator plant with paper towel. Wet your fingertip in the sap and scrub 2-3
leaves of an indicator plant.
3.3.6. Start each scrubbing from the petiole and until the end of the leaf. Avoid excess pressure. Perform
the same procedure for each of the 11 indicator plants.
3.3.7. Immediately after inoculation, wash the sap from the inoculated leaves with a gentle water wash
(using a wash bottle).
3.3.8. Remove all used material, clean the table. Disinfect the gloves (on hands, by washing in bleach 3%
and rinsing in tap water.). Dry hands with paper before proceeding with the next inoculation.

3.4.1. Healthy and positive controls are used for each batch of plants to be evaluated. Healthy and positive
controls are inoculated with sap from healthy and virus-infected plants, respectively. Positive control
includes the following viruses: PVX, PVY, PVS, APLV and APMV.
3.4.2. First inoculate negative controls (healthy plants) and at the end of the process the positive controls.
3.4.3. Virus-infected plants (virus source) are grown in a greenhouse 7 at 18-22°C with 10.000 lux =
0,9290304 lumen/ft2, 15.000 lux. = 1,393456 lumen/ft2. Healthy plants are grown in an isolated greenhouse
in the same conditions as above.
3.4.4. The batch of virus-source plants used to inoculate positive controls are renewed every month and are
given by virology unit (HQU).

Plants showing symptoms are marked with a red stake in the plant pot. The symptoms are registered in a
evaluation summary sheet (Example:Link), after the results are transferred through wireless LAN using a
hand held computer (pocket PC).
Results indicating infection with one virus or more can be visualized in the CIP-VIR system. Results are
notified to the client within the three weeks after finishing the test.

After indicator plants are evaluated, plants must be eliminated placing them into autoclaving plastics bags
size 21 x 40 x 4 cm, during 1 hour at 85°C in the vapor sterilization chamber and after that eliminate the

INTRODUCCIÓN
Los cultivos de papa son susceptibles a la infección de diferentes virus, que pueden dar lugar a una
reducción del rendimiento y calidad de los tubérculos. Generalmente la infección bajo condiciones in vivo
puede ser detectada por la presencia de síntomas en las hojas, tubérculos y el desarrollo de la planta.
Sin embargo, a veces los síntomas no se manifiestan debido a factores tales como la concentración del
virus en la planta, el clima o las reacciones interactivas entre el virus y la planta.
Para determinar si una planta está infectada por un virus, se utilizan los métodos serológicos (ejm.
ELISA, NASH) o moleculares (ejm. PCR). Además, la detección de síntomas en plantas indicadoras es
también un método que puede ser usado. En el CIP, un conjunto de 11 plantas indicadoras se usan
sistemáticamente para detectar el virus de la papa. Este conjunto de plantas indicadoras es conocido por
incluir al menos un huésped susceptible a cada uno de los virus que se transmiten mecánicamente en la
papa. Por analogía, cualquier otro virus desconocido que pueda estar presente en la papa bajo
indexación, tendrá una alta probabilidad de detectarse mediante este procedimiento.
ALCANCE
Algunas especies de plantas reaccionan con síntomas muy visibles y característicos cuando están
infectados con virus de vegetales, entre ellas el cultivo de papa y son las siguientes: Gomphrena globosa
muy susceptible a (PVX) que muestran los síntomas a los 5 días después de la inoculación,y a partir de
los 10 hasta los 15 días en las siguientes plantas como: Nicotiana glutinosa, Nicotina.benthamiana ,
Nicotiana debneyii , Datura stramonium , Nicotiana.tabacum “ White burley ” Chenpodium quinoa ,
Chenopodium murale , Physalis floridana y Lycopersicon esculentum. Estas plantas pueden ser usadas
como plantas indicadoras de infección de virus.
SEGURIDAD
Desinfectar el invernadero 1 semana antes y despues de trabajar un lote nuevo de materiales (ver
punto 3.3).
Usar mandil de laboratorio.
Lavarse las manos antes y después de ingresar al invernadero.
Las plantas indicadoras deberían ser cultivadas en las condiciones mas optimas como sea posible
para estimular el crecimiento rápido y frondoso.
Buen control de insectos vectores de virus (Ejemplo mosca blanca y afidos) dentro del
invernadero.
Usar mascarilla y guantes descartables durante el proceso de inoculación.

PROCEDIMIENTO
1.2.1. En el invernadero, coloque 60 jiffys # 7 en una bandeja de plástico de 24 x 31 x 6 cm, hidratarlos con
4 litros de bencil-dimetilalquil-amonio (Dimanin ®) 0,5 ml/ l. Dejar remojar hasta que se hidraten y hacer un
orificio pequeño, con un diámetro aproximado de un lápiz, en la parte superior del centro de cada jiffy.
1.2.2. Con la ayuda de dos pinzas, saque una plántula de cada tubo de ensayo y extraiga el medio de
cultivo adherido a las raíces. Sea muy cuidadoso en no dañar a las raíces.
1.2.3. Trasplante la plántula en el jiffy de 4.3 cm (3M) e identifíque la con la etiqueta correspondiente
pegada en un estaca.
1.2.4. Esterilice las pinzas con la ayuda de un mechero después de cada trasplante.
1.2.5. Complete el trasplante en la bandeja y agregue un poco de agua destilada a cada plántula para
prevenir la deshidratación.
1.2.6. Cubra la bandeja con una bolsa plástica perforada por el medio, y pegarla con cinta adhesiva a la
bandeja.
1.2.7. Retire el plástico después de 2-3 días.
1.2.8. Coloque la bandeja en una mesa en el invernadero.

2.1.1. Las semillas de las plantas indicadoras son germinadas en bandejas plásticas limpias de 24 x 31 x 6
cm, con seis agujeros en la base para facilitar el drenaje del agua. Las bandejas se desinfectan
previamente con lejía al 3%.
2.1.2. Llenar dos tercios de la bandeja con tierra para la cama de las semillas (Apéndice 2), hacer 10
surcos de 0.5 cm de profundidad y 3-4 cm de distancia entre surco.
2.1.3. Use una bandeja para cada dos especies de plantas indicadoras y coloque 200 semillas (0.2 g
/especie) por surco, se realiza la siembra de las especies indicadoras según los días necesarios para la
germinación, edad ideal o el tamaño de las plantas para la inoculación (ver Tabla 1). Por ejemplo: Orden 1
para Gomphrena globosa; Orden 2 para Nicotiana debneyii, N. glutinosa, etc. ; Orden 3 para Nicotiana
benthamiana; Orden 4 para Chenopodium murale, C. quinoa, etc.
2.1.4. Incubar las bandejas que contienen las semillas a una temperatura de 24-26 °C hasta la emergencia
de las plántulas. Regar con ayuda de un rociador para mantener la tierra húmeda de las bandejas. El
tiempo de germinación de las semillas es de 4-12 días (Ver Tabla 1).

3.3. Inoculación
3.3.1. Solo cuando las plantas indicadoras han sido inspeccionadas y encontradas listas para la
inoculación, se imprime la etiqueta con código de barras para cada accesión. Cuando se imprime la
etiqueta pegarla en una estaca de plástico.
3.3.2. Ponerse los guantes de látex antes de la inoculación.
3.3.3. Esparcir el polvo de carborundun (malla de 600) sobre dos hojas de cada una de las plantas
indicadoras.
3.3.4. Colocar cada bolsa de muestreo que contiene la savia de extracción delante de cada conjunto de
host range, colocando la estaca con la identificación de accesiones en la primera planta indicadora de cada
rango de hospedantes, respectivamente.
3.3.5. Mantener levemente la hoja de la planta indicadora con papel toalla. Moje la yema de los dedos en la
savia y frote 2-3 hojas de la planta indicadora.
3.3.6. Comience cada uno frotando desde el pecíolo hasta el final a la punta de la hoja. Evitar presión
excesiva. Realice el mismo procedimiento para cada una de las 11 plantas indicadoras.
3.3.7. Inmediatamente después de la inoculación, lave la savia de las hojas inoculadas con un lavado de
agua suave (usando una pizeta).
3.3.8. Eliminar todo material usado, limpiar la mesa. Desinfectar los guantes (con las manos, lavándolos en
3% de lejía y enjuagarlos con agua del caño.). Secar las manos con papel antes de proceder con la
siguiente inoculación.

3.4.4. El lote de plantas de fuente de virus usadas para inocular los controles positivos se renuevan todos
los meses y son proporcionadas por la unidad de virologia (HQU).

Paca de 108 litros (3.8 cu ft) compactada. SOGEMIX SM-2 es un sustrato de cultivo a base de musgo
esfágnico y que contiene Vermiculita y Perlita para el cultivo de una gran variedad de especies vegetales.
Este medio de cultivo es muy completo, ligero y uniforme. Ha sido preparado con un alto grado de
consistencia para asegurar el desarrollo vegetal óptimo. Contiene Calcita y Dolomita como corrector de pH
y una carga inicial de fertilizante = Peso = 27-34 kg
* Agua caño 15 litros
* Fertilizante Compuesto (NPK) 20-20-20: 2.5 kg
* Fungicida: Farmathe: (Benomil) = 20 gr
* PCNB: (Pentacloro Nitrobenceno) = 20 gr
* Agua de caño 15 litros
* Combustible: Petróleo Diesel 2 = 12 galones
* Preparación de la arena
* Lavar la arena varias veces con agua de caño con el fin de eliminar sales y residuos en una carretilla un
día antes de la preparación del sustrato
* Trasladar la arena con una carretilla hacia el área de preparación de sustrato
Procedimiento de preparación
Para evaluar 250 entradas por la prueba de rango, preparar 1,250 kg de sustrato, pesar los materiales
según a la tabla 4 como musgo molido, humus de lombriz, y arena de rio lavada. Realizando el proceso en
dos partes debido a que la capacidad de la maquina mezcladora de 650 kg. Colocarlos en carretilla para
trasladar los materiales hacia el área de preparación de sustrato y según el siguiente proceso:
# Desinfección de la maquina mezcladora previo a la introducción del sustrato con legía (hipoclorito de
sodio) al 5 %
# Verter el musgo molido poco a poco en la maquina mezcladora
# Luego el humus de lombriz
# En seguida la arena de rio lavada
# Poner en funcionamiento la maquina mezcladora por 20 minutos con el fin de homogenizar la mezcla del

# Al cabo de los 20 minutos de mezcla, abrir la compuerta inferior de la maquina mezcladora en forma
circular, recepcionar la mezcla en una carretilla para trasladar al coche de des infestación
# Verter el sustrato en el coche de des infestación y humedecer el sustrato con 5 litros de agua corriente
utilizando una regadera, cubrir y cerrar herméticamente con un toldo de lona plastificada, sujetarlo con
cordones de nylon de tal manera que no quede aberturas y evitar la fuga del vapor.
# Trasladar el coche hacia el área de des infestación
# Conectar el coche conteniendo el sustrato a través de una manguera impulsadora de alta presión de 4” a
la maquina impulsadora de vapor de agua.
# En este paso un personal de la unidad de apoyo a la investigación se encarga de poner funcionamiento la
máquina de des infestación.
# Tiempo des infestación 3 horas y a 75oC
# A cabo de este tiempo el coche con el sustrato dejar enfriar por 48 -72horas
# Después del enfriamiento y desinfestado el sustrato trasladar hacia la maquina mezcladora para
incorporar otros materiales como indica en el paso 14
# Poner en funcionamiento nuevamente la maquina mezcladora por 10 minutos y durante este tiempo
incorporar lo siguiente material: Sogemix SM-2+fertilizante compuesto (20-20-20) +Fungicidas: Farmathe y
PCNB, según indica en la tabla 4.
# Después de la mezcla total del sustrato se abre la compuerta inferior de la maquina mezcladora y se
procede a recibir el sustrato en tacho de plástico de 100 litros previamente desinfectado con legía al 2 %
# Almacenar el sustrato en los tacho de plástico, en un ambiente del invernadero 9, previamente
desinfectado con fungicida (Farmathe 2.0 gr/lt), Insecticida: Vertimec 1.5 m/lt, Confidor 1.5 ml/lt y Movento
1.0 gr/lt) y herméticamente cerrados los tachos, con una etiqueta de identificación y fecha de preparación.
# Cerrar la compuerta y apagar la maquina mezcladora
# Unas vez culminado la esterilizacion se procede a la desinfección de la maquina mezcladora y los
materiales usados en este proceso
